赤雹根总皂苷对Ⅱ型胶原性关节炎大鼠IL-17、IL-11表达的影响 被引量:1

The effect of Saponins of Thladiantha Dubia Root on expressions of IL-17 and IL-11 in rats with arthritis induced by type Ⅱ Collagen

摘要 目的:观察赤雹根总皂苷对Ⅱ型胶原诱导性关节炎(Collagen-induced arthritis,CIA)模型大鼠血清及脾脏中白介素-17(Interleukin-17,IL-17)、白介素-11(Interleukin-11,IL-11)表达水平的影响。方法:80只雄性Wistar大鼠,随机取10只作为正常对照组,其余大鼠制备CIA模型。将模型制备成功大鼠随机分为赤雹根总皂苷20、40、80 mg/kg组、雷公藤多苷阳性组和模型组,每组10只,灌胃给予相应药物,连续35d。观察各组大鼠足爪肿胀程度,计算大鼠关节炎指数(AI值);苏木精-伊红染色法(hematoxylineosin staining,HE)观察各组大鼠组织病变;酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)法测定大鼠血清中IL-17、IL-11的蛋白含量;蛋白质印迹(Western Blot)法测定大鼠脾脏中IL-17、IL-11蛋白水平的表达。结果:在给药第35d,与模型组比较,赤雹根总皂苷各剂量组和雷公藤多苷组AI值显著降低;赤雹根总皂苷20mg/kg组、雷公藤多苷组及正常对照组滑膜细胞的数量和炎性细胞数量明显少于模型组;与模型组比较,赤雹根总皂苷20 mg/kg组、雷公藤多苷组大鼠血清IL-17含量明显降低,IL-11含量明显升高;与模型组比较,赤雹根总皂苷各剂量组与雷公藤组大鼠脾脏中IL-17蛋白表达显著降低,赤雹根总皂苷20 mg/kg组与雷公藤多苷组IL-11的蛋白表达显著升高。结论:赤雹根总皂苷对大鼠CIA有明显治疗作用,其治疗机制可能与抑制IL-17的表达,促进IL-11的表达有关。 Objective: To observe the effect of total saponin of Thladiantha dubia root(TSTR) on contents of interleukin-17 and interleukin-11 in serum and spleen tissues in type Ⅱ collagen-induced arthritis(CIA) rats and explore the mechanism of TSTR in treating CIA rats. Methods:10 male Wistar rats were randomly selected in 80 rats as the control group,and CIA models were established in the rest of rats. The model rats were randomly divided into 20,40,80 mg/kg TSTR groups,Tripterygium Glycoside(TG) group and the model group,with 10 rats in each group. All rats were given the corresponding drugs by gavage for 35 d. Then AI values were calculated,lesion tissues of CIA rats were observed by hematoxylin eosin staining(HE),contents of IL-17 and IL-11 in serum of rats were detected by enzyme linked immunosorbent assay(ELISA),and expressions of IL-17,IL-11 protein in spleens of rats were detected by Western Blot assay. Results: Compared with the model group,AI values in TSTR groups and TG group were significantly decreased on thirty-fifth days after the administration of TSTR(p 0. 01). Contents of serum IL-17 in 20 mg/kg TSTR-group and TG group obviously decreased(P〈0. 05),and the content of IL-11 obviously increased(P〈0. 05). Compared with the model group,expressions of IL-17 protein in spleens in TSTR groups and TG group significantly decreased(P〈0. 05 or P〈0. 01),and protein expressions of IL-11 in 20 mg/kg TSTR group and TG group significantly increased(P〈0.05). Conclusion: TSTR has obvious therapeutic effect on CIA in rats,and its therapeutic mechanism may be related to inhibiting the expression of IL-17 and promoting the expression of IL-11.
