摘要
目的:揭示中药淫羊藿(仙灵脾颗粒)及主要成分及代谢产物淫羊藿素调控脂肪肝细胞模型脂代谢的功能,以期获得具有调控肝脏脂代谢的小分子单体药物,为新药研发提供依据。方法:前期动物实验发现淫羊藿苷可以降低肥胖老年大鼠的体重,通过油酸钠处理人肝癌细胞HepG2,构建体外脂肪肝细胞模型;用不同浓度仙灵脾颗粒、淫羊藿苷、宝藿苷Ⅰ、淫羊藿素分别处理模型细胞24h,油红O染色观察不同药物对模型细胞脂质代谢的影响。结果:10μM淫羊藿素处理脂肪肝模型细胞能加强其脂质代谢,减少脂肪肝模型细胞内的脂质聚集作用。仙灵脾颗粒、淫羊藿苷、宝藿苷Ⅰ对脂肪肝细胞模型的脂质代谢未见明显影响,也不影响模型细胞内脂质聚集。结论:中药淫羊藿主要成分的代谢产物淫羊藿素可能具有调控脂质代谢和预防脂肪肝的功能。
Objective: To observe functions of main components of Epimedium and their metabolite epimedium mediating the lipid metabolism on the cell model of the hepatic adipose infiltration,obtain small single molecule drugs which regulate the lipid metabolism in the liver,and provide the basis for the development of new drugs. Methods: The in vitro cell model of the hepatic adipose infiltration was established with human liver cancer cell HepG2 treated sodium oleate. Oil red O hematoxylin staining were used to quantify the lipid accumulation in cells after treated with different concentrations of Icariin,Baohuoside Ⅰ and Epimedium. Results: 10 μM Icariin enhanced lipid metabolism in the hepatic adipose infiltration model cells,and decreased the lipid accumulation. Epimedium,Icariin and baohuoside Ⅰ showed few effect on lipid metabolism in the cell model,no effect of preventing intracellular lipid aggregation. Conclusion: Icariin,a major component's metabolite of Epimedium,may has activities of mediating lipid metabolism and preventing hepatic adipose infiltration.
