免疫组库研究及其在中医药领域应用前景展望

Study on immune repertoires and prospect its application in Traditional Chinese Medicine

免疫组库是指在任何指定时间,某个个体的循环系统中所有功能多样性B细胞和T细胞的总和,反映机体整体免疫能力状态。在免疫组库研究中,利用扩增子补救多重PCR结合高通量测序技术,全面解析T淋巴细胞β肽链或B淋巴细胞免疫球蛋白重链的决定互补区CDR3的多样性,系统地以组学思维解析免疫调节机制。中医药的特色是以系统性思维阐述人体发病机理,重视通过调节免疫功能的平衡实现抵抗力的增强。中医药与现代免疫学研究相结合形成的中医药免疫理论与方法涵盖亚健康及重大疾病的防治。本文旨在通过介绍现代免疫组库技术最新进展,探讨免疫组库研究在中医病因学、证候学、体质学和药物研发等方面的应用前景,可为创新中医药研究内涵提供参考。

The immune repertoires refers to the sum of B cells and T cells with functional diversity in the circulatory system of an individual at any given time,which reflects the state of overall immunity of the body. In the immune repertoires research progression,the diversity of CDR3 in the complementarity-determining region of T lymphocyte beta peptide chain or B lymphocyte immunoglobulin heavy chain was analyzed comprehensively by amplicon rescued multiplex PCR combined with high-throughput sequencing,thus,it can inspect the immuno-regulation mechanism systemically with omics perspective. The systemic demonstration for the pathogenesis of body and emphasis on immune function by adjusting the balance to achieve increased immunity is the characteristic of Traditional Chinese Medicine（ TCM）. Integrating with the modern immunology research,TCM forms the immune theory and methods of its own,which guide the sub-health management and intervention of severe illness. Through introducing the latest advance technology of immune repertoires,we prospect the immune repertoires research on the etiology,symptomatology,constitutional theory and drug discovery of TCM,which can provide a reference for innovating essence of TCM.
