摘要
目的研究10-羟基喜树碱(10-Hcdroxycamptotbecine,10-HCPT)对类风湿关节炎滑膜成纤维细胞(Rheumatoid Arthritis synovial fibroblasts,RASF)基质金属蛋白酶(matrix metalloproteinase-3,MMP3)m RNA表达的影响。方法选取北京大学人民医院2016年8月—2017年1月行全膝关节置换术或关节镜滑膜切除术的类风湿关节炎患者的关节滑膜组织,体外分离培养RASF,用不同浓度的10-HCPT及MTX(1.0、10.0μg/m L)处理RASF,分别为10-HCPT、MTX低、高浓度组,不加药物的细胞作为对照组,培养24 h后,提取细胞RNA,采用荧光定量PCR方法(RT-PCR)检测MMP3-m RNA表达。结果与对照组比较,10-HCPT及MTX高浓度组MMP 3 m RNA表达降低(P<0.01)。对比10-HCPT低浓度组,10-HCPT和MTX高浓度MMP 3 m RNA表达同时降低(P<0.01)。10-HCPT及MTX同浓度组间比较,差异无统计学意义(P>0.05)。结论 10-HCPT可抑制RASF MMP-3 m RNA表达。
Objective To observe the effect of 10-hydroxy-camptothecine(10-HCPT) on m RNA expression level of matrix metalloproteinase-3(MMP-3) in rheumatoid arthritis synovial fibroblasts(RASF). Methods Chose between August 2016 and January 2016 Beijing University People's Hospital of total knee replacement or arthroscopic synovectomy articular synovial tissue in patients with rheumatoid arthritis,RASFs were isolated from synovial tissue of RA patients and cultured in vitro,and then treated with 10-HCPT and Methotrexate(MTX) at high and low concentrations(1. 0 and 10. 0 μg/m L). RASFs without drug intervention were set up as a control group. After 24 h culture RNA was isolated. m RNA expression of MMP-3 was measured by real time PCR(RT-PCR). Results Compared with the control group,m RNA expression of MMP-3 markedly decreased in high concentration 10-HCPT and MTX groups(P〈0. 01). Compared with low concentration 10-HCPT group,m RNA expression of MMP-3 obviously decreased in high concentration 10-HCPT and MTX groups(P〈0. 01). But there was no statistical difference between 10-HCPT groups and MTX groups at the same concentration(P〉0. 05). Conclusion 10-HCPT could inhibit m RNA expression of MMP-3 in RASFs.
