摘要
目的用非田鼠型及田鼠型鼠疫菌分别感染人外周血淋巴细胞,观察高倍显微镜下宿主的敏感性和病理改变。方法分离人外周血淋巴细胞,常规计数铺板培养,用不同型别鼠疫菌感染细胞,MOI为10左右,观察高倍镜下细胞病理变化。结果提取的淋巴细胞总量为2×106个/ml(共76 ml),细胞铺板2 ml/孔,12孔/板×3板;非田鼠型鼠疫强毒菌标准株141活菌计数平均为78个菌,感染指数MOI为9.75;田鼠型鼠疫菌201株平均为39.7个菌,感染指数MOI为9.85。结论淋巴细胞对非田鼠型和田鼠型鼠疫菌均表现出高敏感性,而损害程度依感染菌株型别不同而有较大差别,从病理表型方面验证了非田鼠型鼠疫菌对人的强致病性。
Objective To observe the pathological change of human peripheral blood lymphocytes( PBL) infected with non microtus fuscus Yersinia pestis and microtus fuscus Yersinia pestis respectively. Methods PBL were isolated,counted and carried out on cell plating. Then they were infected with different types of Yersinia pestis at a MOI of about 10. The last step was to observe the pathological change of the cell under the microscope with high magnification. Results The total amount of lymphocytes extracted was 2 × 10^6 cells/ml( 76 ml total),and the cells were plated with 2 ml/well,12 wells/plate × 3 plates. The standard strains of non microtus fuscus Yersinia pestis 141 with strong virulence were 78 cfu/ml,with the MOI being 9. 75; while the strains of microtus fuscus Yersinia pestis 201 were 39. 7 cfu/ml,with the MOI being 9. 85. Conclusion Lymphocytes showed high sensitivity to both non microtus fuscus Yersinia pestis and microtus fuscus Yersinia pestis,but the degree of damage varied from one type of infected strain to another,which proved,from the angle of pathological phenotypes,that non microtus fuscus Yersinia pestis has a stronger pathogenicity.
