摘要
目的建立乐安眠茶水溶性成分的HPLC指纹图谱,测定水溶性成分中斯皮诺素和腺苷的含量及不同泡茶时间二者的溶出度。方法采用HPLC法,色谱条件:Kromasil C18柱(250mm×4.6mm,5μm);流动相为乙腈-0.85mL·L-1磷酸水溶液(5∶95),梯度洗脱;流速为1.0mL·min^(-1);检测波长为260nm;柱温为35℃。结果通过中药色谱指纹图谱相似度评价系统(2004A版),确定了乐安眠茶的HPLC指纹图谱共有模式,标定了31个共有峰,整体相似度为0.993~0.997;10批制剂中斯皮诺素和腺苷的含量与RSD值分别为0.083 4和0.059 1mg·g-1以及1.22%和1.13%;乐安眠茶中斯皮诺素随着浸泡时间的延长,含量逐渐升高,120min时达到峰值;腺苷的含量于水浸泡30min时达到峰值。结论 HPLC指纹图谱法与斯皮诺素、腺苷的含量测定方法以及溶出度的测定方法简便、稳定、科学,可用于乐安眠茶的质量控制。
Objective To establish HPLC fingerprint of Leanmian Tea,to determine the contents of spinosin and adenosine in Leanmian Tea water-soluble part,and to study the dissolution rate of spinosin and adenosine in different soaking time.Methods An HPLC method was used,with Kromasil C18 as column(250 mm×4.6 mm,5μm),acetonitrile-0.85 mL·L^-1 phosphoric acid solution(5∶95)as mobile phase in a gradient elution mode.The flow rate was 1.0 mL·min^-1.The detection wavelength was 260 nm,and the column temperature was at 35 ℃.Results The common mode of HPLC fingerprint for Leanmian Tea was established with Similarity Evaluation System for Chromatographic Fingerprint of TCM(2004 Aedition),among which 31 common peaks were marked.The overall similarity was 0.993-0.997.The contents and RSD of spinosin and adenosine in Leanmian Tea was 0.083 4 and 0.059 1 mg·g-1,1.22% and 1.13%in 10 batches.The content of spinosin was increased over time,and reached peak at 120 min;and the content of adenosine reached peak at 30 min.Conclusion The HPLC fingerprint analysis and the methods of spinosin and adenosine content and dissolution determination are simple,stable and scientific,and can be used for the quality control of Leanmian Tea.
