摘要 目的建立HILIC-HPLC双波长法测定山药中尿囊素和腺苷的含量。方法采用Waters XBridge Amide色谱柱(250mm×4.6mm,3.5μm);以乙腈-水(90∶10)为流动相,体积流量为0.5mL·min^(-1),柱温为30℃;双波长检测为224和260nm。结果尿囊素质量浓度在20.12~319.36μg·mL^(-1)范围内线性关系良好(r=0.999 3),平均加样回收率为93.31%(n=6),RSD值为2.31%;腺苷质量浓度在3.09~49.12μg·mL-1范围内线性关系良好(r=0.999 4),平均加样回收率为90.45%(n=6),RSD值为2.50%。结论该法灵敏、准确、专属性强,可用于山药的质量控制。 Objective To establish an HILIC-double-wavelength HPLC method for the determination of allantoin and adenosine in Rhizoma dioscoreae.Methods Waters XBridge Amide column(250 mm×4.6 mm,3.5μm)was used with acetonitrile and water(90∶10)as the mobile phase in linear gradient elution.The flow rate was 0.5 mL·min^-1,the column temperature was 30℃and double-wavelength were 224 and 260 nm.Results The linear range of allantoin was 20.12-319.36μg·mL^-1(r=0.999 3),the average recovery rate was 93.31%(n=6),RSD was 2.31%;the linear range of adenosine was 3.09-49.12μg·mL^-1(r=0.999 4),the average recovery rate was 90.45%(n=6),RSD was 2.50%.Conclusion The method is sensitive,reliable and accurate,and can be applied in the quality control of Rhizoma dioscoreae.
