摘要
目的探讨高丽槐素对L-02、HEK293、VSMC和H9C2细胞的毒性作用,并初步研究其相关机制。方法采用MTT法检测,凋亡影响用HOE33342和TUNEL法,考察细胞生长抑制率及酶活性。结果高丽槐素对H9C2和L-02细胞增殖和凋亡无影响,L-02细胞对谷草转氨酶(AST)活性降低,对乳酸脱氢酶(LDH)和谷丙转氨酶(ALT)无活性;H9C2细胞无酶活性;对HEK293和VSMC细胞抑制增殖,促进凋亡,LDH活性升高。结论初步确定了高丽槐素对心肌和肝细胞的生长无影响,对人肾上皮细胞和血管平滑肌细胞生长有影响。
Objective To investigate the cytotoxicity of maackiain on L-02,HEK293,VSMC and H9 C2 cells,and to disclose the preliminary study on its mechanism.Methods The inhibition rate of cells growth was detected by MTT method.The apoptosis was investigated by HOE33342 and TUNEL methods,and the enzyme activity was measured simutenously.Results Maackiain had no effect on the proliferation and apoptosis of L-02 and H9 C2 cells;L-02 cells decreased AST activity and was inactive to LDH and ALT.Meanwhile,H9 C2 cells had no enzymatic activity.HEK293 and VSMC cells inhibited the proliferation and promoted the apoptosis,while increased the LDH activity.Conclusion Maackiain did not influence myocardial cells and liver cells,but influenced the human renal epithelial cells and vascular smooth muscle cells.
