摘要
为充分利用东北地区适应性牡丹品种凤丹白进行杂交育种,采用离体萌发法研究其花粉活力,比较蔗糖及H_2BO_3浓度对花粉萌发率的影响,并在适宜培养浓度条件下观察花粉的萌发动态。结果表明:在凤丹白花粉离体萌发试验中硼酸与蔗糖有着重要的影响,0.05g·L^(-1) H_2BO_3+90g·L^(-1)蔗糖浓度培养基为凤丹白最佳花粉培养基,花粉萌发率可达73.7%,花粉萌发出现高峰时段在离体培养的2h后,因培养基浓度不同出现的时间也不相同。
In order to improve the hybrid efficiency of Paeonia ostii Fengdanbai,apeony variety widely planted in northeast area,we used thein vitroculture method to research its pollen vigor,compared the influence of sucrose and boron acid concentrations on pollen germination rate and observed the germinating dynamics.The results showed that boron acid and sucrose played important role on the pollen germination of Fengdanbai.The maximum germination rate of pollen reached 73.7% on the culture supplied with H2BO30.05 g·L(-1) and sucrose 90 g·L(-1).Under this condition,the germination peaks in vitro culture appeared after 2 hours,and the time of peaks occurence was varied with the concentrations of H2BO3 and sucrose in culture medium.
