超声法提取武当丹参药材中蛋白质的正交试验研究

摘要 目的优选武当丹参药材蛋白质的最佳提取工艺,为武当丹参药材蛋白质含量测定提供依据。方法对武当丹参药材蛋白质提取条件料液比、p H值、提取温度、提取时间进行单因素考察,并采用L9(34)正交试验设计法,考察4个因素对蛋白质提取率的影响,从而确定武当丹参药材蛋白质的提取工艺的最优化条件。结果建立了武当丹参药材蛋白质的最优化提取工艺:料液比(武当丹参∶自制纯化水)为1∶50,p H值为12,提取温度为40℃,提取时间设置为20 min。根据该工艺测定武当丹参含可溶性蛋白质的量为10.65%,相对标准偏差为3.74%。结论优化的武当丹参蛋白提取工艺稳定,工艺合理,该提取工艺具有可操作性,能正确反映武当丹参药材中的蛋白质含量。 Objective To optimize the optimum extraction technology of Wudang radix salviae miltiorrhizae protein,and to provide the basis for the determination of protein content of radix salviae miltiorrhizae.Methods Single factor investiga-tion was carried out on the ratio of extraction conditions,p H value,extraction temperature and extraction time of the extractionconditions of Wudang radix salviae miltiorrhizae,and the effect of 4 factors on the extraction rate of protein was investigated bythe orthogonal design method of L9(34),so as to determine the optimum conditions for the extraction process of Wudang radixsalviae miltiorrhiza protein.Results The optimum extraction process of Wudang radix salviae miltiorrhizae protein was estab-lished:the ratio of material to liquid was 1∶50,p H value was 12,the temperature was 40 ℃,and the extraction time was set to20 min.According to the process,the content of soluble protein was 10.65% and the relative standard deviation was 3.74%.Conclusion The optimized extraction process is stable,reasonable and feasible,and can reflect the protein content of salviamiltiorrhiza.
