摘要
为了提高液相色谱在烟草行业的应用,建立了高效液相色谱法测定烟草中烟碱含量的分析方法。烟末样品经过0.01 mol/L的氢氧化钠溶液萃取,以等体积甲醇和0.02 mol/L磷酸盐缓冲溶液作流动相,在259 nm的波长下分析,用外标法定量。结果表明,烟碱含量在0~350 mg/L浓度范围内,线性关系良好,R^2=0.999 1,回收率为95.7%~103.2%。该法具有快速、简单、准确、灵敏等特点,适用于样品的批量检测。
In order to improve the application of high performance liquid chromatography(HPLC) in tobacco industry,a high performance liquidchromatography method was established to determine the content of nicotine in tobacco.The sample was extracted by 0.01 mol/L sodium hydroxidesolution,and the equal volume of methanol and 0.02 mol/L phosphate buffer solution were used for the flow phase,the external standard was quantifiedat 259 nm wavelength.The results showed that the content of nicotine was in the range of 0-350 mg/L,the linear relationship was good,R2=0.999 1,andthe recovery rate was 95.7%-103.2%.The method is fast,simple,accurate,sensitive and so on,which is suitable for bulk detection of samples.
