摘要
目的探究甲酰肽受体1在膀胱癌细胞中的表达及其激活后的作用。方法利用实时荧光定量PCR方法和蛋白免疫印迹法检测正常膀胱上皮细胞和T24和pumc-91细胞甲酰肽受体1mRNA和蛋白的表达量,用甲酰肽受体1的激活剂f MLP激活甲酰肽受体1后,实时荧光定量PCR方法检测IL-8 mRNA表达量。结果基因水平上,甲酰肽受体1在两株膀胱癌细胞和正常的膀胱上皮细胞中均表达,与正常的膀胱上皮细胞相比T24和pumc-91细胞均低表达;蛋白水平上,FPR1在两株膀胱癌细胞中也均表达,与正常膀胱上皮相比T24和pumc-91细胞均高表达。f MLP激活FPR1后,T24和pumc-91膀胱癌细胞IL-8mRNA的表达量增加。结论 FPR1在膀胱癌细胞中高表达并且调控T24和pumc-91细胞IL-8mRNA的分泌,可能与膀胱癌的发生发展相关。
Objective This study was to explore the differential expression and functional role of Formyl Peptide Receptor1 in bladder cancer. Methods The expressions of FPR1 in T24 and pumc-91 cells were measured by qRT-PCR and Western Blot. T24 and pumc-91 cells were cultured with fMLP for 48 h,and then IL-8 mRNA was measured by qRT-PCR. Results FPR1 gene had low expression,while FPR1 protein was over expressed in T24 and pumc-91 bladder cancer cells compared with SV-HUC-1 normal bladder epithelial cells. FPR1 activation improved the expression of IL-8 mRNA. Conclusion FPR1 is highly expressed in bladder cancer cells and regulates the secretion of IL-8 mRNA in T24 and pumc-91 cells,which may be related with the development and progression of bladder cancer.
