白兰氏虫草鸡精对大鼠外周生物钟的调控作用研究 被引量:1

摘要 白兰氏虫草鸡精(Brand’s Essence of Chicken with Cordyceps,BECC)是一种具有增强体质功效的功能性食品,因其能够改善睡眠状况、增强免疫力、缓解疲劳而得到人们的喜爱。为探索BECC在生物节律调节方面的功效,本实验以Wistar大鼠为研究对象,通过颠倒明暗光周期和喂食周期构建倒时差(jet-lag)模型,同时外源性补充BECC,采用红外探头监测大鼠每日行为活动,荧光定量PCR分析外周组织(肝脏和心脏)中生物钟基因转录水平。研究表明:倒时差大鼠服用BECC后,加快了外周组织肝脏和心脏中生物钟基因(Bmal1和Cry1)的重置和昼夜行为活动量的恢复,从而使机体更快地适应时差反应。由此可见,BECC对大鼠外周生物钟系统具有一定的调控作用。 Brand's Essence of Chicken with Cordyceps (BECC) is a kind of dietary supplement which aims to improve sleep quality and fatigue, strengthen the immune system, and enhance physical fitness. To investigate the effects of BECC on the mammalian circadian clock, male Wistar rats were used to establish an ani- mal model of jet lag by reversing both light/dark cycle and feeding schedule, and BECC was exogenously supplemented in their food. Daily locomotor activity was monitored by infrared probes and the mRNA ex- pressions of clock genes in peripheral tissues, including liver and heart, were assessed by real-time quanti- tative PCR. The results indicated that the administration of BECC accelerated the resetting processes of main clock genes, such as Bmall and Cryl in the liver and heart of jet-lagged rats. Locomotor activity of rats recovered quickly, and this contributed to the earlier adaption of jet lag. Therefore, BECC could regulate the peripheral circadian clock in rats.
