化瘀消斑汤对颈动脉粥样硬化兔模型脂代谢及CRP、MMP-9的影响

The Influence of Decoction for Removing Stagnation and Macula on Blood Lipid Level and CRP and MMP-9 of Rabbit Model with Carotid Atherosclerosis

目的:观察化瘀消斑汤对颈动脉粥样硬化兔模型血脂、C反应蛋白(C reactive protein,CRP)、基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)表达及动脉粥样硬化进展的影响。方法:将40只新西兰大耳兔随机分为空白组5只、模型组15只、化瘀消斑汤组10只和阿托伐他汀钙组10只。除空白组外各组大耳兔均给予高脂饲料及静注牛血清白蛋白造模,空白组及模型组给予生理盐水灌胃;化瘀消斑汤组每日予化瘀消斑汤16.35 m L·kg^(-1)灌胃;阿托伐他汀钙组予以阿托伐他汀钙片灌胃,阿托伐他汀钙片研磨成粉状,换算成大耳兔用量按1.09 mg·kg^(-1)标准灌胃。用ELISA法测定血清中血脂水平、CRP及MMP-9含量,光镜下观察病理切片标本。结果:与空白组比较,模型组、化瘀消斑汤组、阿托伐他汀钙组血脂水平、CRP及MMP-9含量明显升高,差异具有统计学意义(P<0.05);与模型组比较,化瘀消斑汤组、阿托伐他汀钙组血脂水平、CRP及MMP-9含量明显降低,差异具有统计学意义(P<0.05);化瘀消斑汤组与阿托伐他汀钙组血脂水平、CRP及MMP-9含量比较,差异无统计学意义(P>0.05)。结论:化瘀消斑汤可改善颈动脉粥样硬化兔血脂水平,减少炎性因子CRP、MMP-9的表达,进而干预颈动脉粥样硬化的进展。

Objective：To observe the influence of Decoction for Removing Stagnation and Macula on blood lipid level and C reactive protein（CRP） and matrix metalloproteinase-9（MMP-9） of rabbit model with carotid atherosclerosis and progress of atherosclerosis（AS） of rabbits with carotid atherosclerosis（CAS）.Methods：Forty New Zealand rabbits were randomly divided into the blank group with 5 cases,15 the model group with 15 cases,Decoction for Removing Stagnation and Macula（DRSM） group with10 cases,and the atorvastatin group with 10 cases.Except the blank group,the ones in other groups were all given high fat diet and intravenous injection of bovine serum albumin for establishing the models.The ones in the blank group and the model group were given normal saline for gavage.The ones in the DRSM group were given DRSM,16.35 m L·kg^（-1） for gavage each day.The ones in the atorvastatin group were given atorvastatin for gavage.Atorvastatin Calcium Tablets were ground into powder and converted into rabbits＇ dosage according to standard of 1.09 mg·kg^（-1） for gavage.Then the serum levels of blood lipids,CRP and MMP-9 were measured by ELISA,and the pathological sections were observed under light microscope.Results：Compared with the blank group,the serum lipid levels,CRP and MMP-9 of the model group,the DRSM group and the atorvastatin group were significantly increased,and all the differences were statistically significant（P〈0.05）.Compared with the model group,the serum lipid levels,CRP and MMP-9 of the DRSM group and the atorvastatin group were significantly decreased,and all the differences were statistically significant（P〈0.05）.The differences in the serum lipid levels,CRP and MMP-9 between the DRSM group and the atorvastatin group were not statistically significant（P〈0.05）.Conclusion：DRSM can improve blood lipid level of rabbit model with carotid atherosclerosis,reduce the expression of inflammatory factors CRP and MMP-9,and then interfere with the progress of carotid atherosclerosis.
