扶正解毒方对脓毒血症大鼠免疫系统调节作用的研究 被引量:7

摘要 目的探讨扶正解毒方对脓毒症大鼠免疫系统的调节作用。方法一次性给予昆明小鼠最大灌胃量2000mg/kg扶正解毒方煎剂,以评价其急性毒性。同时以结扎盲肠穿孔术复制脓毒症大鼠模型,将SD大鼠随机分为对照组、模型组、扶正解毒方组(20mL/kg)、乌司他丁组(1万U/kg),连续给药5 d后检测大鼠血清中白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)、白细胞、中性粒细胞、降钙素原(PCT)、C反应蛋白(CRP)含量以及CD细胞亚群变化。结果扶正解毒方煎剂具有广泛的安全性,安全剂量可高达2000mg/(kg·d)。与对照组大鼠比较,模型组大鼠血清中IL-6、IL-10、TNF-α、白细胞及中性粒细胞、PCT、CRP水平均显著升高,CD4+淋巴细胞百分比、CD4+/CD8+比值显著下降,差异均具有统计学意义(P<0.05)。分别给予脓毒症大鼠扶正解毒方煎剂与乌司他丁后,上述指标均得到显著逆转,与模型组大鼠比较,差异均具有统计学意义(P<0.05)。结论扶正解复方可有效抑制脓毒症大鼠血清中IL-6、IL-10、TNF-α等炎症因子水平、以及白细胞、中性粒细胞计数、降低PCT、CRP含量,有效调节细胞免疫水平,有利于免疫平衡的恢复。 Objective: To investigate the regulating effect of Fuzheng Jiedu Decoction (FZJD) on the immune system of sepsis rats. Methods: 2000 mg/kg FZJD decoction were intragastric administrated once in Kunming mice to evaluate the acute toxicity. Cecal ligation and puncture sepsis model was reproduced in rats,and the SD rats were randomly divided into the control group, the model group, FZJD group (20 mL/kg) and ulinastatin group (10 thousand unit/kg). After administered FZJD detection and ulinastation continuously for five days, interleukin- 6 ( IL -6 ), interleukin - 10 (IL - 10 ), tumor ne c rosis factor alpha ( TNF -α ), white blood c ells, neutrophils, procalci- tonin (PCT) and C-reactive protein (CRP) content and CD cell subsets were detected. Results: FZJD decoction had a wide range of safety with the dose as high as 2000 mg/(kg.d). Compared with the control group,the con- tents of IL-6,IL-10,TNF-α,white blood cells, neutrophils,PCT and C-CRP levels were significantly increased, while the percentage of CD4+ lymphocytes and the ratio of CD4+/CD8+ decreased significantly;the differences were all statistically significant(P〈0.05). After giving septic rats Fuzheng Jiedu Decoction and ulinastatin,the indexes above were significantly reversed. Compared with the model group,the difference was statistically significant (P〈 0.05 ). Conclusion: Fuzheng Jiedu Decoction can effectively inhibit the levels of inflammatory factors such as IL- 6, IL-10 and TNF-α in the serum of septic rats,as well as leukocyte,neutrophil count,reduce the content of PCT and CRP, and effectively regulate cellular immune level, which is conducive to the recovery of immune balance.
