摘要
目的探讨视神经损伤后再生的动物模型的建立方法。方法实验研究Wistar健康大鼠64只,随机分为3组:正常组(4只),无任何处理;对照组(20只),完全剪断视神经并对位缝合;实验组(40只),人为造成外伤性白内障外余操作同对照组。处死动物后,在蓝色激发光的照射下,用低倍荧光显微镜观察大鼠整个视路的荧光情况。结果对照组大鼠视神经无视神经轴突再生,罗丹明-B异硫氰酸盐荧光完全被阻于视神经近眼球断端。实验组34只大鼠视神经出现明显视神经轴突再生,大量罗丹明-B异硫氰酸盐荧光突破视神经断端。对照组与实验组之间再生成功率相比,差异有统计学意义(r=39.231,P=0.000)。结论视神经完全横断性损伤联合晶状体损伤的实验大鼠可以成功地作为一种较好的研究视神经损伤后再生的动物模型。
Objective To investigate the methods of establishing animal model of regeneration of optic nerves after section injury. Methods A experimental study. Sixty-four Wistar healthy rats were divided into 3 groups randomly:normal group of 4 rats, no treatment was given; control group of 20 rats, the optic nerve of rats was completely sectioned and appositionally sutured; experimental group of 40 rats, received the same dispose with control group and traumatic cataract were formed artificially. After killing animals, the entire visual pathways were observed under blue light by low power fluorescence microscope. Results The rhodamine-B-isothiocyanate fluorescence was stopped by optic nerve blockage in control group with no axon regeneration. The obvious regeneration of optic nerve was observed in experimental group and rhodamine-B-isothiocyanate fluorescence was penetrated the optic nerve blockage. Conclusion It is a better animal model of optic nerves regeneration for the sectioned optic nerves of rats with traumatic cataract.
