摘要
目的观察白藜芦醇对过氧化氢诱导人视网膜色素上皮细胞凋亡和细胞周期的影响。方法培养的人RPE细胞系分为正常对照组、过氧化氢损伤组和白藜芦醇保护组,过氧化氢损伤组加入200 mg/L过氧化氢,白藜芦醇保护组加入50 mg/L白藜芦醇作用24 h后加入200μmol/L过氧化氢培养2、8、24 h。采用CCK-8检测细胞抑制率,流式细胞术检测细胞周期和细胞凋亡。结果 CCK-8法检测发现白藜芦醇保护组明显降低了细胞抑制率,且与氧化损伤组相比差异有统计学意义(P<0.05)。流式细胞术检测结果显示经白藜芦醇保护作用2、8、24 h后,细胞凋亡率明显降低,差异有统计学意义(P<0.01)。处于G0/G1期细胞比例和氧化损伤组相比明显降低,差异有统计学意义(P<0.01);处于S期细胞比例和氧化损伤组相比明显升高,差异有统计学意义(P<0.01)。结论白藜芦醇对过氧化氢诱导人视网膜色素上皮细胞凋亡有一定的抑制作用,其效应可能和细胞周期调节有关。
Objective To observe the effects of resveratrol on human retinal pigment epithelium cellsagainst H_2O_2-induced cell apoptosis and cell cycle. Methods Human RPE cells were divided into 3 groups ran-domly including normal control group,H_2O_2 injury group and resveratrol protected group. H_2O_2 injury group wastreated with 200 μmol/L H_2O_2,and resveratrol protected group were treated with 50 mg/L resveratrol and then treat-ed with 200 μmol/L H_2O_2. CCK-8 was used to observe inhibitory rate of cell,and the flow cytometry was applied todetect the apoptosis and cell cycle distribution. Results CCK-8 assay showed that the inhibitory rate of cell treat-ed with resveratrol(50 mg/L)were obviously decreased,which was significantly different with H_2O_2 injury group(P〈0.05),the apoptosis levels of cell treated with resveratrol for 2,8,24 h were significantly decreased(P〈0.01). Moreover,the cell cycle distribution in the G0/G1 phases was also significantly decreased(P〈0.01),thecell cycle distribution in the S phases was significantly increased(P〈0.01). Conclusion Resveratrol can inhibitthe cell apoptosis on human retinal pigment epithelium cells induced by H_2O_2,which is related to cell cycle regula-tion.
