电针对阿尔兹海默病大鼠海马区M2型小胶质细胞极化的影响 被引量:18

Study on electroacupuncture promoting the polarization of M2 phenotype microglia cells in hippocampus of AD model rats

摘要 目的:探讨促进海马小胶质细胞向M2极化是否为电针治疗阿尔兹海默病(AD)的作用机制。方法:通过侧脑室注射Aβ制作老年痴呆大鼠模型,采用形态学和分子学方法,研究电针治疗AD调控小胶质细胞极化的机制。结果:电针能降低AD模型大鼠Aβ的沉积,促进大鼠海马区M2型小胶质细胞(Iba1+Arg1+)的生成(P<0.01),抑制M1型小胶质细胞(Iba1+i NOS+)的生成(P<0.01),促进大鼠海马组织Arg1蛋白的表达(P<0.01),抑制i NOS蛋白的表达(P<0.01),抑制大鼠海马组织细胞因子IL-1β和TNF-αm RNA的表达(P<0.01)。结论:电针百会穴可调控大鼠海马区小胶质细胞极化状态,促进小胶质细胞向M2型方向极化,抑制M1型小胶质细胞的生成,起到改善大鼠AD病变的效果。 Objective: To explore the mechanism of electroacupuncture on Alzheimer's disease, whether by promoting the polarization of microglia cells in hippocampus to M2 phenotype or not. Methods: The AD rat model was established by injection of Aβ in the lateral ventricle. Morphological and molecular methods were used to study the mechanism of electroacupuncture on the polarization of microglia cells. Results: Electroacupuncture significantly reduce the deposition of Aβ, and increased the number of M2 phenotype microglia cells(Iba1+Arg1+) in hippocampus(P〈0.01), inhibited the information of type-M1 microglia cells(Iba1+i NOS+)(P〈0.01), and up-regulated the protein expression level of Arg1 in hippocampus(P〈0.01), down-regulated the protein expression level of i NOS(P〈0.01). And the m RNA expression levels of cytokine IL-1β and TNF-αwere down-regulated(P〈0.01). Conclusion: Electroacupuncture at the acupoint of Baihui(DU16) has effects on rats with Alzheimer's disease, by regulating polarization of microglia cells in hippocampus, promoting the polarization of microglia cells to M2 phenotype and inhibiting the information of M1 phenotype microglia cells.
