双龙方对大鼠肠系膜上动脉环舒缩功能的影响及机制探究 被引量:1

Effects and mechanism of Shuanglong Formular on the contractile function of the superior mesenteric artery in rats

摘要 目的:研究双龙方对大鼠离体肠系膜动脉血管舒缩功能的影响,探讨其可能存在的机制。方法:应用离体微血管动力描记技术和离子通道特异性阻断方法,考察双龙方对大鼠肠系膜动脉环直接舒张作用,及对受体依赖型、电压依赖型钙通道功能的影响,和钾离子通道抑制剂对双龙方舒张血管作用的影响。结果:双龙方可舒张大鼠肠系膜上动脉,32、16、8g生药/L组与空白对照组比较有显著性差异(P<0.01)。双龙方8g生药/L组5-HT最大收缩达到KCl预收缩高度(Emax),与空白对照组比较显著降低(P<0.01)。双龙方8、4、2g生药/L浓度组中,NA的Emax较空白对照组显著下降(P<0.01,P<0.05)。双龙方8、4g生药/L组KCl Emax较空白对照组均显著降低(P<0.01)。双龙方8g生药/L组Ca Cl2 Emax较空白对照组显著降低(P<0.01)。4种类型钾离子通道抑制剂4-氨基吡啶、氯化钡、格列苯脲、四乙胺干预后,双龙方20、10g生药/L组血管的舒张幅度与空白对照组比较,无显著性差异。结论:双龙方有显著的扩张血管作用,其舒张血管的作用机制与抑制电压依赖型钙离子通道功能及5-羟色胺、去甲肾上腺素受体有关。 Objective: To investigate the effects of Shuanglong Formular on the contractile function of mesenteric artery in rats and the potential mechanism responsible for it. Methods: The microvascular dynamic mapping technique and ion channel specific blocking method was applied to investigate the effects of Shuanglong Formular on the direct relaxation of mesenteric artery ring in rats and on the function of receptor-dependent and voltage-dependent calcium channels, and the effect of potassium channel inhibitor on the vasodilation of Shuanglong Formular was investigated. Results: Shuanglong Formular could relax the superior mesenteric artery, showing significant differences(P〈0.01) between 32 g/L, 16 g/L and 8 g/L groups and the blank control group. The maximum shrinkage of 5-HT in Shuanglong Formular 8 g/L group reached KCl preshrunk height(Emax), and it was significantly lower than the blank control group(P〈0.01). In Shuanglong Formular 8 g/L, 4 g/L and 2 g/L groups, the Emax of NA was significantly lower than that in the blank control group(P〈0.01, P〈0.05). The KCl Emax Shuanglong Formular 8 g/L and 4 g/L groups were significantly reduced compared with the blank control group(P〈0.01). In Shuanglong Formular 8 g/L group, Ca Cl2 Emax was significantly reduced compared with the blank control group(P〈0.01). After the intervention of four types of potassium channel inhibitors 4-AP, Ba Cl2, Glib and TEA, the diastolic amplitude of the blood vessels of Shuanglong Formular 20 g/L and 10 g/L groups showed no significant difference with the blank control group. Conclusion: Shuanglong Formular can significantly dilate blood vessels. The underlying mechanism is related to inhibition of voltage dependent calcium channel function and 5-HT and NA receptors.
