有氧运动对糖尿病前期人群血浆代谢物的影响 被引量:18

Effects of Aerobic Exercise on Plasma Metabolites in Prediabetes Subjects

摘要 目的:探讨有氧运动对糖尿病前期(PDM)人群血浆代谢物的影响。方法:选择25名PDM受试者,以及与之年龄、性别匹配的血糖正常和新发2型糖尿病(T2DM)受试者各10名。对PDM受试者分别进行为期12周的健康教育(n=12)和有氧运动干预(n=13),有氧运动干预方案:40%~60%储备摄氧量,每周3次,每次50分钟。采用液相色谱质谱(LC-MS)技术分析血浆代谢产物谱,通过主成分分析法研究不同血糖受试者之间、有氧运动干预前后PDM受试者血浆代谢产物谱的差异。结果:与干预前相比,运动组PDM受试者在干预后的体脂百分比、餐后2小时血糖、低密度脂蛋白胆固醇均明显降低(平均下降4.6%、16.22%和9.27%)。比较运动前后PDM受试者的血浆代谢物,发现31种差异性代谢物,其中磷脂酰胆碱、溶血磷脂胆碱、鞘磷脂、甜菜碱等25种代谢物含量上调,油酸、胞嘧啶等6种代谢物下调。结论:有氧运动对PDM患者血浆代谢物调节作用显著,磷脂酰胆碱、溶血磷脂胆碱、鞘磷脂和油酸等物质可能是有氧运动改善PDM患者糖脂代谢的潜在生物学标志性。这些发现可帮助我们更好的理解有氧运动预防T2DM的作用机制。 Objective To examine the effect of aerobic exercise training on plasma metabolites inpre-diabetes mellitus(PDM)subjects. Methods Twenty-five PDM subjects were selected,and each 10 age-and sex-matched with normoglycemia and new-onset T2 DM subjects were enrolled. PDM subjectsunderwent aerobic exercise(n=13)or health education(n=12)for 12 weeks. The aerobic exercise was con-ducted 3 times/week,50 min/time at 40%-60% of oxygen uptake reserve. Liquid chromatography-massspectrometry(LC-MS)was used to analyze the spectrum of plasma metabolites. Principal component anal-ysis was employed to study the differences in plasma metabolite profiles between subjects with differentglycemic level and between PDM subjects before and after aerobic exercise. Results After training,thebody fat percentage,2-hour plasma glucose,and low density lipoprotein cholesterol of PDM subjects re-duced significantly(by 4.6%,16.22% and 9.27% on average). Comparison of plasma metabolizers inPDM subjects before and after exercise revealed 31 differential metabolites,with 25 metabolites such asphosphatidylcholine,lysophosphatidylcholines,sphingomyelin and betaine increasing,and 6 metabolites,such as oleic acid and cytosine decreasing. Conclusion Aerobic exercise has a marked effect on theplasma metabolites in PDM subjects. Phosphatidylcholine,lysophosphatidylcholines,sphingomyelin and oleic acid may be the potential biomarkers of aerobic exercise improving the glycolipid metabolism inPDM subjects. These findings can help us better understand the mechanism of aerobic exercise prevent-ing T2 DM diabetes.
