摘要
目的:观察苦参碱对结肠癌SW480细胞凋亡的影响,并分析其作用机制。方法:使用苦参碱对结肠癌SW480细胞进行处理,作为实验组;同时选择空白组作为对照组。采用噻唑蓝比色法检测苦参碱对细胞增殖的作用,Hoechst检测对细胞凋亡的作用,流式细胞术检测细胞凋亡率,蛋白免疫印迹法检测苦参碱对蛋白表达的影响。结果:培养24 h、48 h、72h后,实验组SW480细胞均出现明显的生长抑制,并随着药物质量浓度的提高和时间的延长而增强。对照组SW480细胞凋亡率为(4.26±0.25)%,明显低于实验组的(9.85±1.03)%、(15.62±1.98)%、(19.69±2.86)%、(26.58±3.58)%、(33.95±5.26)%;SW480细胞经过不同质量浓度的苦参碱作用48 h后出现了明显的细胞凋亡,并且随着质量浓度的提高,细胞凋亡率增加(P<0.05)。与对照组比较,实验组Bcl-2含量降低,Bax含量升高(P<0.05);实验组各组中p-Akt含量均明显降低(P<0.05),总Akt含量无明显变化(P>0.05)。结论:苦参碱能够通过抑制Akt信号通路,抑制SW480细胞生长,并且能够诱导细胞发生凋亡。
Objective: To investigate the effect of matrine on apoptosis of colon cancer SW480 cells induced by Akt signaling pathway and to investigate its mechanism. Methods: Matrine treatment of colon cancer SW480 cells as the experimental group,and blank group as control group,using MTT colorimetric assay to detect the effect on cell proliferation,Hoechst detection of cell apoptosis,flow cytometry detection of cell apoptosis rate,Western blot detection of the effect of matrine on protein expression. Results:After cultured with 24 h,48 h and 72 h,the SW480 cells in the experimental group showed marked inhibition of growth and increased with the increase of the concentration of drugs and the prolongation of time. The apoptosis rate of SW480 cells in the control group was( 4. 26 ± 0. 25) %,significantly lower than the experimental group( 9. 85 ± 1. 03) % and( 15. 62 ± 1. 98) % and( 19. 69 ± 2. 86) % and( 26. 58 ± 3. 58) % and( 33. 95 ± 5. 26) %. SW480 cells treated with matrine at different concentrations of 48 h showed that there was significant cell apoptosis,and with the increase of the concentration,the apoptosis rate increased( P 〈0. 05). Compared with the control group,the content of Bcl-2 in the experimental group decreased,and the content of Bax increased( P〈 0. 05). In the experimental groups,the content of p-Akt decreased significantly( P〈0. 05),and there was no significant change in the total Akt( P〉 0. 05). Conclusion: Matrine can inhibit the growth of SW480 cells by inhibiting the Akt signaling pathway and induce apoptosis in the cells.
