天王补心丹联合迷迭香精油对阿尔兹海默病模型小鼠记忆功能的影响

Intervention Effect on Alzheimer's Disease in Mice by Tianwang Buxin Dan Combined with Rosemary Essential Oil

目的:观察天王补心丹联合迷迭香精油对阿尔兹海默病模型小鼠记忆功能的影响。方法:50只SD小鼠随机平均分为5组:空白组、模型组、吸入组、中药组、中药+吸入组。除空白组外,其余各组分别腹腔注射东莨菪碱3 mg,1次·d^(-1),连续14 d。从第15天开始,吸入组吸入迷迭香精油,每次30 min,3次·d^(-1);中药组灌胃天王补心丹4.71g·kg^(-1),2次·d^(-1);中药+吸入组联合吸入组和中药组的方法,共8 d,空白组和模型组给予等体积生理盐水。于给药前1 d,给药后1 d、4 d、8 d,T迷宫测试各组小鼠的记忆能力,旷场实验测试各组小鼠的跨越格数和潜伏期;末次给药1 h后,分析主要脏器系数。结果:给药1 d后,中药+吸入组跨越格数[(32.78±2.96)个]显著高于模型组[(23.15±1.30)个](P<0.05);中药+吸入组潜伏期[(2.91±0.19)s]显著低于模型组[(4.24±0.51)s](P<0.05)。中药+吸入组T迷宫记忆测试正确率(36.26%)显著高于模型组(17.91%)(P<0.05);给药4 d后,中药+吸入组跨越格数[(40.56±3.08)个]显著高于模型组[(25.13±1.71)个](P<0.05);中药+吸入组潜伏期[(2.54±0.21)s]显著低于模型组[(4.19±0.42)s](P<0.05)。给药8 d后,吸入组T迷宫记忆测试正确率(38.01%)、中药组(39.78%)和中药+吸入组(45.21%)均显著高于模型组(18.22%)(P<0.05);吸入组跨越格数[(41.37±3.16)个]、中药组[(39.46±3.14)个]、中药+吸入组[(51.22±4.24)个]显著高于模型组[(24.68±1.49)个](P<0.05);中药+吸入组潜伏期[(1.97±0.06)s]显著低于模型组[(4.18±0.39)s]。中药组心脏系数(3.938 9±0.131 2)和肝脏系数(0.384 6±0.055 8)均显著低于模型组心脏系数(4.401 5±0.391 4)和肝脏系数(0.540 8±0.083 4)(P<0.05)。结论:天王补心丹和迷迭香精油均可改善阿尔兹海默病小鼠的记忆功能,但以二者联合使用效果为佳;迷迭香精油联合天王补心丹对小鼠记忆力有改善作用,可以兴奋中枢神经系统,使其在新异环境中的自发活动和探究行为明显增加;天王补心丹对阿尔兹海默病模型小鼠有一定的不良反应,迷迭香精油可降低天王补心丹对阿尔兹海默病模型小鼠的不良反应。

Objective： To investigate the intervention effect on alzheimer＇s disease in mice using Chinese medicine Tianwang Buxin Dan combined with rosemary essential oil. Methods： A total of 50 SD mice were divided into 5 groups randomly. In addition to the normal group,each group received scopolamine（ 3 mg·kg-1,one time a day） by intraperitoneal injection for 14 days. From the 15 th day,the inhaling group inhaled Rosmarinus officinalis essential oil（ 30 min/time,3 times/d） and the Chinese medicine group received Tianwang Buxin Dan by gavage（ 4. 71 g·kg-1,2 times/d）. The combined medication group used the methods of inhalation and Chinese medicine for 8 days. The normal control group and model group were treated with physiological saline. T maze tested the memory ability of mice and open field tested the span lattice and latency of mice before medication 1 d and after medication 1 d,4 d,8 d. The last 1 h after the treatment,and main organ coefficients were analyzed. Results： after dosing 1 d,compared with model group,Chinese medicine and inhalation group across the number was（ 32. 78 ± 2. 96） significantly higher than the model group（ 23. 15 ± 1. 30）,which had statistical significance（ P〈 0. 05）,the incubation period of（ 2. 91 ± 0. 19） s significantly shorten than model group（ 4. 24 ± 0. 51） s（ P〈 0. 05）; after dosing 4 d,Chinese medicine ＋ inhalation group T maze memory test accuracy was 36. 26% significantly higher than the model group 17. 91%（ P 〈0. 05）,and the number across was（ 40. 56 ±3. 08） significantly higher than the model group（ 25. 13 ± 1. 71）（ P 〈0. 05）,and the incubation period was（ 2. 54 ± 0. 21） s significantly shorten than that of model group（ 4. 19 ± 0. 42） s. After dosing 8 d,T maze memory test accuracy in inhalation group was 38. 01%,Chinese medicine group was 39. 78% and Chinese medicine ＋ inhaled group was 45. 21%,which were significantly higher than that of model group 18. 22%（ P〈 0. 05）,the number across in inhalation group was（ 41. 37 ± 3. 16） and traditional Chinese medicine group（ 39. 46 ± 3. 14）,Chinese medicine group ＋ inhalation group（ 51. 22 ± 4. 24） significantly higher than the model group（ 24. 68 ± 1. 49）,and the difference was statistical significance（ P〈 0. 05）. The incubation period of TCM ＋ inhalation group was（ 1. 97 ± 0. 06） s significantly shorter than that of model group（ 4. 18 ± 0. 39） s. Heart and liver coefficient in the Chinese medicine group was（ 3. 938 9 ± 0. 131 2） and（ 0. 384 6 ± 0. 055 8）,which was significantly lower than the model group（ 4. 401 5 ± 0. 391 4） and（ 0. 540 8 ± 0. 083 4）,and the difference was statistical significance（ P 〈0. 05）. Conclusion： Tianwang Buxin Dan and rosemary essential oil can improve the function of the memory of alzheimer＇s disease in mice,but the combination of the two effects is preferred; Rosemary essential oil and Tianwang Buxin Dan could improve the memory in mice,can be excited central nervous system,make its spontaneous activities and to explore the behavior in the environment of extraneous increased significantly; Tianwang Buxin Dan has certain side effects on alzheimer＇s disease in model mice,rosemary essential oil can reduce the side effects.
