摘要 通过复合蛋白酶水解青稞蛋白以获得活性多肽,并研究其抗氧化性。运用响应面(RSM)分析对青稞蛋白酶解工艺条件进行优化,在单因素试验基础上,以多肽得率为考察指标,研究底物浓度、酶解温度、酶用量、p H等因素对多肽得率的影响,并通过分析多肽对自由基的清除作用来判断其抗氧化性。结果表明:青稞蛋白酶解最佳工艺条件为:在底物浓度为2%,酶解温度为46℃,酶用量为10000U/g,p H为7.45的条件下酶解3h,青稞多肽得率达10.45%。该研究结果为青稞蛋白的开发利用提供了科学依据。 To optimize enzymolysis technology of highland barley protein to obtain active peptide and determine antioxidant abilities of it. This paper deals with the optimization of process conditions for the enzymatic hydrolysis of highland barley protein using response surface methodology( RSM),the effects of hydrolysis time,temperature,p H,concentration ratio and enzyme dose on peptide yield were investigated based on single factor experiments,the antioxidant abilities of peptide were evaluated by measuring free radicals scavenging ability. The results showed that the optimum conditions of hydrolysis with hydrolysis time is 3 h,temperature is46℃,p H is 7. 45,concentration ratio is 2%,and enzyme dose is 10000 U/g,peptide yield is 10. 45%. the results establish the theory basis for the application of highland barley protein.
