摘要
目的:观察补肾活血法对子宫内膜异位症(EMs)模型大鼠子宫内膜DNA甲基化的影响。方法:取80只造模成功EMs模型大鼠随机分为治疗组、对照组,另取40只大鼠结扎一侧子宫作为空白组。治疗组予补肾活血免煎颗粒,对照组和空白组予生理盐水,灌胃12周后雌雄2:1合笼,各组妊娠大鼠于妊娠第6天尾静脉采血后处死。测定EMs模型大鼠血清E2、P水平,检测子宫内膜HOAX10蛋白表达,检测子宫内膜HOAX10基因DNA甲基化。结果:治疗组EMs模型大鼠E2、P含量与对照组比较明显增高(P<0.05)。治疗组HOXA10分泌期呈强阳性(3+)表达,与对照组比较有统计学意义(P<0.05)。治疗组治疗后HOXA10甲基化率明显降低,与对照组比较有统计学意义(P<0.05)。结论:补肾活血中药可降低EMs模型大鼠HOXA10异常甲基化率,增加HOXA10蛋白表达。
Objective:To observe the effects of Chinese traditional Chinese medicine of nourishing kidney andactivating blood on the DNA methylation of endometriosis (EMs)model rat endometrial homologous box gene(HOXA10) Methods:80 endometriosis model rats were randomLy divided into treatment group and control group,and the other 40 rats tied to one side of the uterus were divided into a blank group The treatment group were givenBushenhuoxue particles ,The control group and blank group were given saline,After 12 weeks both male and female 2:1 cages,The pregnant rats of each group were extracted caudal vein and executed after the 6th day of pregnancy The E2 and P levels of EMs model were measured The expression of the endometrium HOAX10 proteinwere detected The DNA methylation of the endometrium HOAX10 gene were detected Results: The serum EMsmodel of the treatment group E2,P and control group were significantly higher (P 〈 0.05)t The expression ofHOXA 10 in the treatment group was positive (3 + ),which was statistically significant (P 〈 0.05) After treatment,the HOXAl0 methylation rate decreased significantly,which was statistically significant compared with thecontrol group (P 〈 0.05)Conclusion:Traditional Chinese medicine of n nourishing kidney and activating blood could reduce the abnormal methylation rate of HOXAl0 and increase the expression of HOXAl0.
