摘要
目的探讨骨髓内皮祖细胞(endothelial progenitor cells,EPCs)在糖尿病周围神经病变(diabetic peripheral neuropathy,DPN))治疗中的作用机制。方法用链脲佐菌素60mg/kg腹腔注射制备DPN大鼠模型。将雄性SD大鼠分为正常组(NC组)、DPN组、DPN+EPCs组和DPN+生理盐水组(DPN+S组)。检测大鼠坐骨神经运动神经传导速度(motor nerve conduction velocity,MNCV).以免疫组化检测NF.KB和髓鞘碱性蛋白(myelin basic protein,MBP)在坐骨神经的表达水平。结果与DPN组相比,DPN+EPCs组的NF.KB表达明显减少(P〈0.05),DPN+S组的NF—KB表达增加,差异有统计学意义(P〈0.01)。DPN+EPCs组的MBP表达量与DPN组相比差异无统计学差异(P〉0.05),但较DPN+S组增加(P〈0.01)。结论EPCs移植能抑制NF—KB、增加MBP的表达,这可能有利于神经损伤的修复。
Objective To explore the mecha-nism of bone marrow-derived endothelial progenitor cells (EPCs) in the treatment of peripheral neuropathy of diabetic rats. Methods Rats with diabetic peripheral neuropathy (DPN) were induced by streptozotocin ( 60 mg/kg). Male SD rats were divided into normal control group ( NC group), DPN group, DPN + saline group (DPN + S group), and DPN + EPCs group. Sciatic nerve motor nerve conduction velocity (MNCV) was measured. The expressions of NF-KB and myelin basic protein (MBP) in sciatic nerve were detected by immunohistochemistry. Results Compared with DPN group, the expression of NF-KB was reduced in the sciatic nerve of DPN+ EPCs group, while the expression of NF-KB was increased in the sciatic nerve of DPN + S group. There was no statistical difference in the expression of MBP between DPN and DPN + EPCs groups. Compared with DPN + S group, the expression of MBP was higher in DPN + EPCs group. Conclusion Transplantation of EPCs inhibits the expression of NF-KB and increases the expression of MBP, which might be conducive to the repairs of nerve injury.
