摘要
目的:通过检测大建中汤治疗前后脾阳虚证大鼠胃癌组织中的Cox-2、NF-кB mRNA的表达,探讨大建中汤治疗脾阳虚胃癌的部分作用机制,为温阳法治疗肿瘤的新思路提供科学依据。方法:将SD大鼠随机分为正常对照组、模型对照组、西药对照组及大建中汤高、低剂量组,对除正常对照组外的其余组别大鼠进行脾阳虚胃癌模型制备后,正常对照组予0.9%氯化钠注射液灌胃,模型对照组予番泻叶水浸液灌胃,药物组分别给予大建中汤高、低剂量和环磷酰胺干预,于末次给药后处死动物,取胃黏膜异常处胃组织2块,液氮速冻,待测。结果:与正常对照组比较,模型对照组大鼠胃组织中的Cox-2、NF-кB mRNA的含量明显升高(P>0.01);与模型对照组比较,大建中汤高、低剂量组及西药对照组大鼠胃组织中的Cox-2、NF-кB mRNA的含量明显降低(P>0.01或P>0.05),大建中汤高剂量组含量优于低剂量组(P<0.05)。结论:大建中汤可能通过调节Cox-2、NF-кB的含量从而改善脾阳虚胃癌大鼠的症状。
Objective: To investigate the mRNA expression of cyclooxygenase-2(COX-2) and nuclear factor-kappa B(NF-кB) in rats with gastric carcinoma with spleen yang deficiency before and after the treatment with Dajianzhong decoction and the mechanism of action of Dajianzhong decoction in the treatmen t of gastric carcinoma with spleen yang deficiency,and to provide a scientific basis for yang-warming method in tumor treatment. Methods: Sprague-Dawley rats were randomly divided into normal control group,model control group,Western medicine control group,and high-and low-dose Dajianzhong decoction groups. All rats except those in the normal control group were used to establish a model of gastric carcinoma with spleen yang deficiency; then the rats in the normal control group were given 0. 9% sodium chloride injection by gavage,those in the model control group were given the water extract of senna by gavage,and those in the high-and low-dose Dajianzhong decoction groups and the Western medicine control group were given high-and low-dose Dajianzhong decoction and cyclophosphamide,respectively. The rats were sacrificed after the last administration,and two tissue blocks of abnormal gastric mucosa were collected and quickly frozen with liquid nitrogen for future examination. Results: Compared with the normal control group,the model control group had significant increases in the mRNA expression of COX-2 and NF-кB in gastric tissue(P〈0. 01); compared with the model control group,the high-and low-dose Dajianzhong decoction groups and the Western medicine control group had significant reductions in the mRNA expression of COX-2 and NF-кB in gastric tissue(P〈0. 01 or P〈0. 05),and the high-dose Dajianzhong decoction group had significantly greater reductions than the low-dose Dajianzhong decoction group(P〈0. 05). Conclusion: Dajianzhong decoction can improve the symptoms of rats with gastric carcinoma with spleen yang deficiency by regulating the content of COX-2 and NF-кB.
