全反式维甲酸对OCT4、NANOG阳性U87人脑胶质瘤细胞作用的研究

摘要 目的研究全反式维甲酸(ATRA)对OCT4、NANOG阳性U87人脑胶质瘤细胞分化增殖的影响.方法筛选OCT4、NANOG呈阳性的U87人脑胶质瘤细胞,用全反式维甲酸(ATRA)处理细胞后检测细胞增殖、细胞周期和分化相关基因的表达.结果ATRA处理后的细胞中G0/G1期细胞比例显著增加,细胞周期出现G0/G1期阻滞,但ATRA对U87细胞分化无明显作用.结论ATRA通过调控细胞周期的方式影响OCT4、NANOG阳性U87细胞增殖. Objective To examine the effect of all-trans retinoic acid (ATRA) on the proliferation and differentiation of OCT4, NANOG positive U87 human glioma cells. Methods OCT4, NANOG positive cells were isolated from U87 human glioma cells and were treated with ATRA. Then the cell cycle and certain transcription factors were examined. Results The number of cells in G0/G1 phase signifcantly increased after ATRA treatment, indicated that ATRA may exert growth inhibitory effects through the induction of G1 arrest. However, ATRA did not affect the differentiation of U87 cells. Conclusion ATRA may affect the cell proliferation of OCT4, NANOG positive U87 cells through regulating cell cycle.
