摘要
目的:观察姜黄素对七氟醚引起的成年小鼠记忆提取障碍的影响。方法:成年昆明小鼠120只,雌雄各半,随机分为对照组(Con组),七氟醚组(Sev组)和七氟醚加姜黄素组(Cur组);Cur组连续6 d腹腔注射0.3 mg/g的姜黄素,Con组和Sev组注射等体积的二甲基亚砜和生理盐水混合液,之后Sev组、Cur组吸入3.3%七氟醚2 h,Con组吸入40%O_22 h;3组小鼠于吸入前及停止吸入后12、24、48及72 h进行避暗实验,记录各组小鼠5 min内错误次数、步入潜伏期及Δ潜伏期,评价小鼠记忆提取能力。结果:与Con组比较,Sev组吸入七氟醚后12、24 h步入潜伏期缩短,Δ潜伏期负值增大和5 min内错误次数增加(P<0.05);与Sev组比较,Cur组吸入七氟醚后12、24 h步入潜伏期延长,Δ潜伏期负值减小和5 min内错误次数减少(P<0.05);而吸入七氟醚后48、72 h时,Sev组、Cur组步入潜伏期和错误次数比较,差异无统计学意义(P>0.05)。结论:姜黄素预处理可改善七氟醚引起的成年小鼠短期记忆提取障碍。
Objective: To observe the effect of curcumin on sevoflurane-induced memory retrieval dysfunction in adult mice. Methods: Adult Kun Ming mice of 120 with both sexes were randomly divided into three groups: control group( group Con),sevoflurane group( group Sev) and sevoflurane +curcumin group( group Cur). According to the different time of the test,mice in group Cur were made an intraperitoneal injection of curcumin with the dose of 0. 3 mg/g for 6 consecutive days while group Con and Sev were injected into isometric mixed liquids of dimethyl sulfoxide and mormal saline,then group Sev and group Cur were exposed to 3. 3% sevoflurane for 2 h and group Con inhaled 40% O2 for 2 h. The three groups were made step-through test before inhalation and after inhalation at 12 h,24 h,48 h,72 h respectively. Stepping-in the lantency,Δlantency and error times in 5 minutes of all groups were tested to evaluate the capability of memory retrieval in mice. Results: Compared with group Con,group Sev at 12 h and 24 h after inhalation with sevoflurane showed significant differences( P〈0. 05),stepping-in latency was decreased,Δlantency and error times in 5 minutes were increased; group Cur at 12 h and 24 h after inhalation with sevoflurane also showed significant differences( P〈0. 05),stepping-in latency was prolonged and Δlantency was increased compared with group Sev. But there was no significance in statistics comparing the stepping-in lantency with error times of Group Sev and Cur after inhalation with sevoflurane at 48 h and 72 h( P〉0. 05). Conclusion: Pre-administration of Curcumin can improve sevoflurane-induced memory retrevail dysfunction of adult mice.
