摘要
目的:探究瘤周围刺联合环磷酰胺(CTX)对4T1乳腺荷瘤小鼠HSP70及局部免疫的影响。方法:将造模成功的4T1乳腺荷瘤小鼠随机分为模型组、药物组、针刺组、针药组,其中模型组不干预;药物组隔天腹腔注射CTX(50 mg/kg);针刺组予瘤周围刺(1次/d);针药组予腹腔注射CTX配合瘤周围刺。实验干预2周后测瘤质量计算抑瘤率,免疫组化(IHC)染色热休克蛋白(HSP)70后采用IPP6.0图像分析软件计算阳性表达结果,流式细胞术(FCM)检测肿瘤组织中CD4^+CD25^+Treg、PD-L1阳性表达情况。结果:针刺或CTX均可抑制肿瘤生长、降低HSP70表达,针刺联合CTX在抑制肿瘤生长和HSP70表达上具有协同作用。CTX可减少CD4^+CD25^+Treg细胞表达量、增加PD-L1阳性率,但针刺或针药联合对此无明显作用。结论:针刺联合CTX对肿瘤微环境中HSP70表达具有抑制作用,但对Treg细胞、PD-L1无明显作用。
Objective: To study the effects of peritumoral acupuncture combined with cyclophosphamide (CTX) on HSP70 and local immunity in tumor-bearing mice. Methods: The 4TI breast tumor-bearing mice were randomly divided into model group, drug group, acupuncture group and group of acupuncture combined drug. The model group was not intervened; The drug group received intraperitoneal injection with CTX (50 mg/kg) ev- ery other day; The acupuncture group received peritumoral acupuncture treatment every day; The group of acupuncture combined drug received peritumoral acupuncture and intraperitoneal injection with CTX. After 2 weeks of experimental intervention, inhibition rate was observed. HE staining and IPP6.0 were used to observe the expression of HSP70. Flow eytometry was used to detect the expression of PD-LI and CD4+CD25+Treg in tumor tissue. Results: Acupuncture or CTX could inhibit tumor growth and reduce the expression of HSP70. Acupuncture combined with CTX showed synergistic effect on tumor growth and the expression of HSPTO. CTX can reduce the expression of CD4+CD25+Treg cells and increase the positive rate of PD-L1, but acupuncture or acupunc- ture combined showed no significant effect. Conclusion: Acupuncture combined with CTX can inhibit the expression of HSP70 in tumor mieroenvironment, but it shows no obvious effect on the expression of PD--L1 and Treg cells.
