摘要
目的探讨血小板源性生长因子-C(PDGF-C)在视网膜变性小鼠模型的神经保护作用。方法检测在体外条件下PDGF-C的作用。使用的动物模型包括Pde6b^(rd1)小鼠和MNU诱导的视网膜光感受器细胞损伤模型。两种模型均分为两组,在视网膜下分别注射AAV-PDGF-C-Ires-ZsGreen和AAV-Ires-ZsGreen,然后通过切片HE染色观察视网膜光感受器细胞层厚度的变化来检测PDGF-C的神经保护作用。结果在Pde6b^(rd1)小鼠的视网膜下注射AAV-PDGF-C-Ires-ZsGreen,外核层与全层视网膜厚度比为(0.575 918±0.14),较对照组A(0.335 821±0.11)明显增加,差异有统计学意义(P<0.05)。本实验在MNU诱导的视网膜光感受器细胞损伤模型中,注射AAV-PDGF-C-Ires-Zs Green的实验组B(0.239 712±0.030 9)中外核层厚度较对照组B(0.193 552±0.022 4)增厚,差异有统计学意义(P<0.05)。结论 PDGF-C在光感受器细胞损伤的动物模型中具有较强的神经保护作用。
Objective To investigate the neuroprotective effect of platelet derived growth factor-C(PDGF-C)in retina degeneration models. Methods Two animal models had been tested. One was a retinitis pigmentosa model called Pde6b^rd1,another model was MNU-induced photoreceptor lesion mouse model. The two models were divided into two groups respectively:subretinal injection of AAV-PDGF-C-Ires-Zs Green and AAV-Ires-ZsGreen. Neuroprotective effect of PDGF-C was assessed by HE staining. Results Subretina injection of AAV-PDGF-C-Ires-ZsGreen in Pde6b^rd1 mice increased the outer nuclear layer to(0.575 918±0.14),while only(0.335 821±0.11)in AAV-Ires-ZsGreen A group(P〈0.05). Furthermore,the neuroprotective effect was also detected in MNU-lesioned photoreceptor loss mouse model. Thickness of outer nuclear layer in AAV-PDGF-C-Ires-ZsGreen B group(0.239 712±0.030 9)was higher than that in AAV-Ires-Zs Green B group(0.193 552±0.022 4)(P〈0.05). Conclusion PDGF-C has a strong neuroprotective effect in animal models of photoreceptor cell injury.
