摘要
旨在研究GlyCAM-1在绵羊自然发情子宫组织中的表达变化,以期为周期发情绵羊子宫中GlyCAM-1的表达受孕酮诱导调控的推测提供支持。运用实时荧光定量PCR和免疫印迹法对绵羊自然发情后子宫第10、12、14、16和18天GlyCAM-1 mRNA和蛋白表达变化进行检测。实时荧光定量PCR结果显示:在绵羊发情后的第10~18天之间,子宫组织中GlyCAM-1的表达呈现先下降再上升的趋势,发情后的第10天GlyCAM-1在转录水平表达出现峰值,并且显著的高于另外4个时段(P<0.05),第12天GlyCAM-1 mRNA水平显著下降(P<0.05),第14~16天的表达量保持相对恒定,第18天GlyCAM-1 mRNA水平再次显著升高(P<0.05),但仍低于第10天水平(P<0.05)。Western Blotting检测结果显示:绵羊在自然发情后的第10~18天,子宫内膜上的GlyCAM-1蛋白表达量的变化亦呈现出先下降后上升的趋势,与其他4个时间点相比第10天的蛋白表达量达到峰值(P<0.05),第12~16天GlyCAM-1的蛋白表达量没有显著变化(P>0.05),GlyCAM-1的蛋白表达量在第18天显著升高(P<0.05)。GlyCAM-1在转录与翻译水平的表达变化一致,呈现先下降后上升的趋势。该结果为周期发情绵羊子宫中GlyCAM-1的表达可能受孕酮诱导调控的推测提供支持。
To study the expression of GlyCAM-1 in uteri of sheep during for the regulation of progesterone induced GlyCAM1 expression in oestrous cycle, and provide support sheep during oestrous cycle, the transcription and translation of GlyCAM-1 on day 10, 12, 14, and 18 of oestrous cycle were respectively detected by RT-PCR and Western transcription and translation levels of blotting. The results showed that from day 10 to day 18, both the GlyCAM-1 in uteri first decreased and then increased, with the peaks on day 10, which were significantly higher than those on other days. GlyCAM-1 mRNA level significantly decreased on day 12, kept relatively stable between day 14 to day 16, and significantly increased on day 18.GlyCAM-1 protein level was stable between day 12 to day 16, significantly higher on day 18 (P〈0.05). However, both the without significant change (P〉0.05), but was mRNA and protein levels on day 18 were significantly lower than those on day 10 (P〈0.05). The transcription level of GlyCAM-1 was consistent with the translation level of GlyCAM-1. The results provide support for the speculation that expression of GlyCAM-1 in uteri of sheep during oestrous cycle may be induced by progesterone.
