抗菌肽等抑菌物质抗菌活性检测方法比较研究 被引量:3

摘要 为了快速测定抑菌物质的抗菌活性,选取大肠杆菌、沙门氏菌和金黄色葡萄球菌为试验菌种,以乳酸链球菌素(Nisin)为试验材料,分别采用抑菌圈法和分光光度法检测Nisin溶液的抗菌活性,建立抑菌圈法和分光光度法的联系。最后对茶多酚溶液和聚赖氨酸溶液的抗菌活性进行检测分析。结果表明:不同浓度的Nisin溶液,其抑菌圈直径比与抑菌率的关系,对大肠杆菌、沙门氏菌和金黄色葡萄球菌分别为指数、三次多项式和二次多项式。Nisin溶液对不同菌种抑菌效果的抑菌圈法与分光光度法之间的关系不同;同一菌种,Nisin溶液抑菌效果的抑菌圈法与分光光度法之间的关系是确定的。因此,对于确定的菌种来说,通过建立Nisin溶液抑菌效果的抑菌圈法与分光光度法之间的关系,可以用分光光度法快速简便的测定Nisin溶液抗菌活性。经验证该方法同样适用于茶多酚和聚赖氨酸等抑菌物质。 In order to rapidly determine the antibacterial activity of antimicrobial substances, Escherichia coli(ATCC25922), Salmonella(LT2) and Staphylococcus aureus(26111) selected as test strains, the relationships of between methods for detecting antimicrobial activity of antimicrobial substances were founded with inhibition zone method and spectrophotometry. The results showed that the relationships between the diameters of the bacteriostatic circle and the inhibition rate were exponential, three times polynomial and two polynomial for Escherichia co1i(ATCC25922), Salmonella(LT2) and Staphylococcus aureus(26111) respectively in different concentrations of Nisin solution. The relationship between inhibition zone method and spectrophotometry was different for the effect of Nisin solution on the Bacteriostasis of different strains and was the same for the same strains. Therefore, spectrophotometry can be used to quickly and easily determine the Nisin solution antibacterial activity for the determination of the specific strains through the establishment of Nisin solution antibacterial effect of the relationship between the inhibition zone method and spectrophotometry. The method is also suitable for tea polyphenols, poly lysine and other antibacterial substances.
