摘要
目的探讨miRNA-3129与hace1是否可作为口腔肿瘤标志物,在口腔肿瘤的治疗过程中发挥作用。方法收集15组口腔肿瘤组织与瘤旁组织,应用QRT-PCR法检测组织中hace1 mRNA的表达,通过MTT实验、细胞划痕实验观察hace1对细胞增殖、迁移的变化影响。生物信息学预测hace1的miRNA,通过luciferase和QRT-PCR验证miRNA-3129与hace1的关系,QRT-PCR法检测组织中miRNA-3129的表达。结果 hace1在口腔肿瘤中低表达,并抑制口腔肿瘤的迁移与增殖;且miRNA-3129可靶向hace1 3’UTR,并调节hace1表达水平。结论 miRNA-3129在口腔肿瘤中高表达,与hace1表达呈负相关性,可能通过靶向hace1促进口腔肿瘤生长。
Objective To investigate whether hace1 and miRNA-3129 can be used as a oral tumor marker and play a role in the treatment of oral tumor. Methods 15 groups of oral tumor tissue and tumor adjacent tissue were collected,and the mRNA level of hace1 was detected by QRT-PCR method. The effects of hace1 on cell proliferation and migration were observed by MTT and cell scratch test. Bioinformatics predicts the miRNA which target to hace1,and the relationship between miRNA-3129 and hace1 was verified by luciferase and QRT-PCR,and the expression of miRNA-3129 in oral tumor tissues was detected by QRT-PCR. Results Hace1 was low expressed in oral tumors,and inhibited migration and proliferation of oral tumors; miRNA-3129 could target hace1 3' UTR and regulate hace1 expression. Conclusion The miRNA-3129 is highly expressed and negatively correlated with hace1 expression in oral tumor,may promote oral tumor growth by targeting hace1.
