摘要
目的:探讨miR-93在多囊卵巢综合征(PCOS)患者血清中的表达及临床意义。方法:利用实时荧光定量PCR分别检测PCOS患者(25例)和正常对照(25例)血清中miR-93的相对表达量,并分析miR-93与PCOS临床特征(高雄激素血症、胰岛素抵抗)的关系;绘制ROC曲线评价血清miR-93诊断PCOS的价值。结果:与对照组相比,miR-93在PCOS患者血清中的相对表达量明显升高(Z=7.614,P<0.01);体质指数(BMI)、空腹血糖(FBG)、血清总睾酮(TT)和胰岛素抵抗指数(HOMA-IR)在PCOS患者中均升高(P<0.05)。在对照组,血清miR-93的水平与TT、HOMA-IR的相关系数r值分别为0.03、0.39(均P>0.05);在PCOS组,血清miR-93的水平与TT、HOMA-IR的相关系数r值分别为0.33、0.28(均P>0.05)。ROC曲线分析显示,血清miR-93用于诊断PCOS的曲线下面积为0.73,敏感度为70%,特异度为78%。结论:miR-93在PCOS患者血清中表达上调,可能与PCOS的发生发展相关,且用于诊断PCOS具有一定的价值。
Objective: To investigate the expression and clinical significance of sermn miR-93 in patients with polycystic oval-/syndrome (PCOS). Methods: The relative expression level of serum miR-93 was detected in PCOS group (n =25) and control group (n =25) by quantitative real time PCR, and the relationship between miR-93 and clinical features of PCOS (byperandrogenemia and insulin resistance) were analyzed. Furthernlore, the value of sermn miR-93 was evaluated in the diagnosis of PCOS using ROC curve. Results: Compared with the control group, the relative levels of serum miR-93 in PCOS patients were significantly increased (Z =7.614, P 〈 0.01), and body mass index (BMI), tasting blood-glucose (FBG), sermn total testosterone (TT) and insulin resistance index were elevated in PCOS patients (P 〈 0.05). In control group, correlation eoetfieient r values of sermn miR-93 with TT and HOMA-IR were 0.03 and 0.39 (both P 〉 0.05), respectively. In PCOS group, correlation eoetfieient r values of sermn miR-93 with TT and HOMA-IR were 0.33 and 0.28 (both P 〉 0.05), respectively. ROC curve analysis showed that the area under the curve, sensitivity and specificity of sermn miR-93 for the diagnosis of PCOS were 0.73, 70%, and 78%, respectively. Conclusion: The expression levels of miR-93 in PCOS patients are up-regulated, therefore the increment of miR-93 may be associated with the occurrence and development of PCOS, and it has certain value for the diagnosis of PCOS.
