摘要
探讨自胚胎时期至出生后持续镧暴露对子代大鼠的肝肾发育状况的影响。Wistar雌性大鼠32只,随机分成对照组及低、中、高镧暴露组,每组8只雌鼠,饮水中氯化镧(LaCl_3)浓度分别为0%、0.25%、0.5%和1.0%,仔鼠在断乳前经由母体血循环和吸吮母乳染镧,断乳后则通过自行饮水摄入。分别于仔鼠出生后14 d、28 d和42 d随机抽取各窝仔鼠1只,每组8只,取肝脏、肾脏称重,计算脏器系数。各剂量组仔鼠出生后体重持续增加,对照组的体重增长趋势最为明显,生后14 d、28 d和42 d各剂量组仔鼠的体重无差异(P>0.05)。生后14 d和28 d各组仔鼠的肝脏重量、肝脏系数无统计学差异(P>0.05);42 d各染镧组肝脏大于对照组肝脏,但未出现统计学差异(P>0.05),0.5%LaCl_3组肝脏系数高于对照组(P<0.05),1.0%LaCl_3组肝脏系数高于其它3组(P<0.05)。在各个生长发育阶段不同剂量组仔鼠肾脏重量无统计学差异(P>0.05);生后14 d和28 d不同剂量组仔鼠的肾脏系数无差异;生后42 d各染镧剂量组仔鼠肾脏系数明显增加,0.25%LaCl_3组肾脏系数高于对照组(P<0.05),0.5%LaCl_3组肾脏系数高于对照组(P<0.05),1.0%LaCl_3组肾脏系数高于其它3组(P<0.05)。由此看出,随着染镧剂量的增加,以及染镧时间的延长,仔鼠的肝脏和肾脏的发育受到了影响,其脏器系数均随染镧剂量增加而增加。
To investigate the effects of lanthanum exposure on the development of liver and kidney of offspring rats.32 Wistar female rats were randomly divided into control group and low,medium and high lanthanum exposed group.The concentration of lanthanum chloride in drinking water was 0%,0.25%,0.5% and1.0%,respectively.Offspring rats exposed to lanthanum by blood circulation of their mother and sucking milk before weaning,and exposed to lanthanum by self-drinking water after weaning.Eight rats from each litters in each groups were randomly selected on the 14 th day,28 th day and 42 nd day after birth.The liver and kidney were weighed and the organ coefficient was calculated.The body weight of the offspring in each dose group increased continuously,and the body weight of the offspring was not different at 14 d,28 d and 42 d after birth(P〉0.05).The liver weight and liver coefficient of the offspring were not statistically different(P〉0.05) at 14 d and 28 d.On the 42 nd da,the weights of liver in lanthanum groups were larger,but there was no statistical difference(P〉0.05),the liver coefficient of 0.5% LaCl3 group was higher than that of the control group(P0.05).and the liver coefficient of 1.0% LaCl3 group was higher than that of the other three groups(P〉0.05).There was no significant difference in the weight of kidneys in different dose groups(P〉0.05).There was no significant difference in the renal coefficient of the offspring from 14 d and 28 d.On the 42 nd day,the renal coefficient of 0.25% LaCl3 group and 0.5% LaCl3 group were all higher than that of the control group(P〉0.05),and the renal coefficient of 1.0% LaCl3 group was significantly higher than that of the other three groups(P0.05).It suggested that the developments of liver and kidney of offsprings were affected by the increasing and the prolongation of lanthanum exposure,and the organ coefficient increased with the increasing of lanthanum dose.
