葡萄砧木品种‘3309M’中microRNA的鉴定

摘要 为了解葡萄砧木microRNA(miRNA)的表达模式,采用高通量测序法对砧木品种‘3309M’根和叶片中的miRNA文库进行分析。结果表明:在葡萄砧木根和叶片文库中,共鉴定了181条保守miRNA和104条新的miRNA,长度以21 nt为主,其5'端首位碱基对U具有很强的偏向性;不同miRNA家族其成员数量也不同,这些miRNA的靶基因功能主要为植物激素信号转导和糖酵解/糖异生等;筛选出显著差异表达的miRNA,其中54条miRNA表达量上调,33条miRNA表达量下调。本研究为认识葡萄砧木特异miRNA及进一步分析其功能提供了基础。 In order to understand the expression pattern of microRna (miRna) in grape rootstocks, high-throughput sequencing was used to analyze the miRna libraries in the roots and leaves of the '3309M' rootstock. The results showed that 181 conserved miRNAs and 104 new miRNAs were identi?ed in the roots and leaves of grape rootstock. We found the length of miRNA mainly was 21 nt, and the ?rst base pair of 5' terminal had strong bias in U, and the number of members of di?erent miRNA family was also di?erent. The functions of target genes of these miRnas were mainly focus on plant hormone signal transduction and glycolysis/gluconeogenesis etc. In additional, 54 up-regulated and 33 down-regulated miRnas were also found. This study provided the basis for understanding speci?c miRNAs of grape rootstocks and further analysis of their functions.
