不同针刺法对C57/BL6小鼠黑素生成、ACTH及相关细胞因子表达的影响

摘要 目的:观察不同针刺法对C57/BL6小鼠黑色素生成、促肾上腺皮质激素(ACTH)及相关细胞因子(IL-1β、TGF-β、TNF-α)表达的影响及作用机制。方法:将48只小鼠随机分为空白组、火针组、普通针刺组及冰针组,肉眼观察小鼠皮肤黑色素生成情况,并行组织学检查,采用免疫组化法检测小鼠皮肤中ACTH、IL-1β、TGF-β、TNF-α的表达。结果:火针组最早出现黑色素生成,至第14天,HE染色中黑色素颗粒明显多于普通针刺组及冰针组。至第3天,各针刺组ACTH表达均高于空白组(t值分别为9.36、6.37、5.87,P值均<0.05),其中火针组表达强于普通针刺组及冰针组(t值分别为3.42、2.93,P值均<0.05)。与之对应,IL-1β在火针组的表达均强于普通针刺组、冰针组,第3天最明显(t值分别为2.88、2.94,P值均<0.05);TGF-β第3天可见火针组表达强于普通针刺组、冰针组,差异有统计学意义(t值分别为3.01、3.45,P值均<0.05)。各组间TNF-α的表达则无明显改变。结论:不同针刺法均明显促进C57/BL6小鼠黑色素生成,上调ACTH、IL-1β及TGF-β的表达,其中以火针组作用最为明显。 Objective: To analyze the function of different accupucture in C57/BL6 mice pigmentogenesis,and detect the expression of ACTH and related cytokines,including IL-1β、TGF-β、TNF-α during the process. Methods: Forty-eight C57/BL6 mice were used as the experiment subjects,certain area of each mouse skin was irradiated with fire needles,ice needles or normal acupuncture respectively. Efficacy was evaluated at the 1 st day,3 rd day,7 th day and 14 th day of the experiment by observation,HE staining,immunohistochemistry to stain ACTH、IL-1β、TGF-β and TNF-α. Results: The expression of ACTH could be detected in the beginning in every needle group,which was obviously higher than that of blank group( t = 9. 36,6. 37,5. 87,P〈0. 05) at the 3 rd day,and the expression of ACTH in fire needle group were higher than common-needle group and ice-needle group( t = 3. 42,2. 93,P〈0. 05). After the fire-needle treated,the expression of IL-1β on the 3 rd day was stronger than common-needle group or ice-needle group( t =2. 88,2. 94,P〈0. 05). The expression of TGF-β is stronger than common-needle group,ice-needle group on the 3 rd day( t = 3. 01,3. 45,P〈0. 05). However,the expression of TNF-α in different periods of different groups had no obvious differences. Conclusion: Different acupuncture methods could increase the expression of ACTH,enhance melanogenesis. The effect of the fire needle group is the most obvious.
