摘要
目的研究重庆市男男性接触(MSM)者中未接受抗病毒治疗的人类免疫缺陷病毒1(HIV-1)感染者的耐药突变发生情况和流行情况。方法收集2014年1月至2015年12月调查的148例HIV-1感染者外周血,提取血浆中的HIV基因组RNA,采用In-house方法扩增PI基因全序列和逆转录酶基因1~250氨基酸编码序列,产物纯化后送诺赛基因公司进行序列测定。序列拼接后,上传至斯坦福大学HIV耐药数据库进行基因耐药性分析。结果 148例标本中,1例存在蛋白酶抑制剂主要耐药突变,2例存在次要突变,14例存在非核苷类逆转录酶抑制剂相关突变。结论在重庆市MSM人群中已存在低流行状态原发性耐药相关基因突变,须加强对该人群的监测。
Objective To investigate the occurrence and prevalence of drug resistance mutations in human immunodeficiency virus(HIV)-1 infected patients who were not treated with antiviral therapy among men who have sex with men(MSM)in Chongqing. Methods The peripheral blood of 148 HIV-1 infected persons surveyed in Chongqing from 2014 to 2015 was collected.HIV genomic RNA was extracted from the patient′s plasma,and the entire PI gene sequence and 1 to 250 amino acid reverse transcriptase gene sequences were amplified using the In-house method.The purified product was sent to Nossa Gene Company for sequence determination. The sequences were spliced and uploaded toStanford University′s HIV drug resistance database for analysis of gene resistance. Results Of the 148 samples,1 had major protease inhibi-tor resistance mutations,2 had minor mutations,and 14 had non-nucleoside reverse transcriptase inhibitor related mutations. Conclusion There is already a prevalence of primary resistance-related gene mutations in the MSM population in Chongqing,and monitoring of this popula-tion must be strengthened.
