虾夷扇贝糖胺聚糖的提取及降血脂活性的研究 被引量:2

摘要 以虾夷扇贝为原料,酶解提取糖胺聚糖,利用响应面法优化虾夷扇贝糖胺聚糖的提取工艺条件。结果表明,试验因素对提取率影响的顺序依次为料液比>酶解时间>酶添加量。回归模型中失拟项的p>0.05,复相关系数为0.998 6,说明模型具有较高的拟合度。最佳提取工艺为酶添加量1.44%,料液比1∶4(g∶m L),酶解时间2.54 h,在此条件下虾夷扇贝糖胺聚糖提取率为1.873%。动物试验结果表明,虾夷扇贝糖胺聚糖中、高剂量组均能显著降低高脂模型小鼠的总胆固醇、甘油三酯、低密度脂蛋白水平;高剂量组能显著提高高密度脂蛋白水平,因此虾夷扇贝糖胺聚糖对高脂模型小鼠具有较好的降血脂作用。 The extraction conditions of glycosaminoglycan from Patinopecten yessoensis by enzymatic hydrolysis were optimized using response surface analysis. The results showed that the order of effects of 3 factors on the extraction rate was in turn solid-liquid ratio〉 enzymolysis time〉 enzyme dosage. In the regression model,the p value in lack of fit 0.05 and the complex correlation coefficient was 0.998 6,showed that the model had a high fitting degree. The optimum extraction technology was confirmed as follows:enzyme dosage 1.44%,the ratio of material to loquid 1∶4(g∶m L),and enzymolysis time 2.54 h.Under this condition,the extraction rate of glycosaminoglycan reached 1.873%. Animal test results showed that glycosaminoglycan from P. yessoensis decreased the contents of total cholesterol, triglyceride and low density lipoprotein-cholesterol in serum and increased high density lipoprotein-cholesterol significantly in the experimental hyperlipidemic mice. These results suggested that glycosaminoglycan from P. yessoensis could modulate the level of blood lipid in hyperlipidemia mice.
