摘要
为了开发利用子二代大鲵黏液,研究了酶解大鲵黏液提取透明质酸(HA)的工艺。通过高效液相色谱法、傅立叶变换红外光谱法、核磁碳谱分析了HA的分子量、所含基团,以及体外清除自由基和还原Fe^(3+)检测了其抗氧化能力。结果表明,影响胰蛋白酶提取大鲵HA的主要因素为酶解温度和酶解时间,大鲵HA分子量为4.85×106 Da,具有体外清除DPPH·,·OH,ABTS^+·和还原Fe^(3+)能力。因此,大鲵黏液可作为透明质酸潜在的来源。
To comprehensively utilize skin mucus from cultured giant salamander(Andrias davidianus),trypsin was used to digest mucus for purification hyaluronic acid(HA). HPLC, FT-IR, and 13 C-NMR were employed to analyze molecular weight and group residue. Antioxidative activity were assayed by scavenging DPPH·,·OH and ABTS^+·,as well as reducing Fe^3+ in vitro. The results showed that the main factors affecting the purification of HA from mucus of A. davidianus were enzymatic hydrolysis temperature and time. HA,with molecular weight of 4.85×10^6 Da,had antioxidation in vitro. The results indicated that mucus of A. davidianus was considered as a potential source of valuable HA.
