粒细胞集落刺激因子受体在急性髓系白血病患者中的表达意义

摘要 目的:研究给予G-CSF治疗后,经化疗后粒细胞数量减少的AML患者粒细胞G-CSFR的表达水平以探寻其临床意义。方法:选取AML患者18例作为观察对象,根据患者治疗情况将其分为初诊组10例、用药组8例。另选取同期正常对照组8例。采用ELISA法测定血浆中G-CSF含量,采用RT-PCR法检测G-CSFR mRNA水平,采用Western blot法检测G-CSFR蛋白表达水平。比较各组血浆G-CSF含量、G-CSFR mRNA和G-CSFR蛋白表达水平。结果:初诊组与对照组比较,G-CSF浓度无统计学差异(P>0.05),G-CSFR mRNA和G-CSFR蛋白表达水平明显降低(P<0.05);用药组与初诊组比较,G-CSFR mRNA和G-CSFR蛋白表达水平均明显升高,但是仍低于对照组(P<0.05)。结论:用G-CSF提高髓系白血病放化疗后的粒细胞数量是安全的,粒细胞表面的G-CSFR表达水平可用于疗效的评定。 Objective: To explore expression of granulocyte colony-stimulating factor( G-CSFR) on AML patients with granulocytopenia after G-CSF treatment and chemotherapy and its significance. Methods: 18 AML patients were selected as the research objects and divided into newly diagnosed group( 10 cases) and medication group( 8 cases). 8 healthy subjects at the same period were chosen as the control group. The plasma G-CSF content was measured by ELISA,the G-CSFR mRNA expression level was tested by RT-PCR,and Western blot was used to detect the protein expression of G-CSFR. Results: There was no difference in the concentration of G-CSF between the newly diagnosed group and the control group( P〈0.05),but the expression of G-CSFR mRNA and G-CSFR decreased( P〈0.05). Compared with the newly diagnosed group,the expression of G-CSFR mRNA and G-CSFR increased,but they were still lower than that in the control group( P〈0.05). Conclusions: It is safe to use G-CSF to improve the number of granulocytes in the AML patients after chemotherapy. The level of G-CSFR expression on the surface of the granulocytes can be used to evaluate the curative effect.
