摘要
目的 探讨大蒜素对硝苯地平致牙龈增生(DGO)患者牙龈成纤维细胞(HGFs)I型胶原分泌和转化生长因子β1(TGF-β1)蛋白表达的影响.方法 对不同牙龈增生(GO)指数患者行龈切术,收集增生牙龈组织.组织块培养法分离培养DGO-HGFs,在培养液中加入不同浓度(0μg/ml、3μg/ml、6μg/ml、9μg/ml)大蒜素,作用72h后取细胞上清液,酶联免疫吸附试验(ELISA)测定Ⅰ型胶原及TGF-β1的含量.结果 不同浓度大蒜素可使不同增生程度DGO-HGFs细胞上清液中I型胶原(P〈0.05)和TGF-β1含量降低(P〈0.05),且大蒜素作用浓度越高、牙龈增生程度越重,两者含量降低越显著.结论 大蒜素可抑制DGO-HGFs I型胶原分泌,此抑制作用与牙龈增生程度、药物浓度相关,其作用机制可能与下调TGF-β1表达有关.
Objective To study the effect of allicin on the secretion of collagen type I and transforming growth factor- β 1 from human gingival fibroblasts in nifedipine-induced gingival overgrowth patients. Methods Gingival fibroblasts were derived from three patients with different levels of nifedipine-induced gingival overgrowth.Cells were induced by 3 p. g/ml, 6 μg/ml, 9μg/ml allicin (low-and mid-and high-concentration drug intervention groups ) , respectively. DGO-HGFs without allicin were served as blank control. Collagen type I and TGF-β1 were checked at 72h after stimulation by enzyme link immunosorbent assay ( ELISA ) . Results The secretion of collagen type I and TGF- β1 expression was decreased by Allicin in certain range in the way of dose-dependent effect and degree of hyperplasia dependent effect ( P〈0.05 ) . Conclusion Allicin can decrease the secretion of collagen from human gingival fibroblast nifedipine-induced gingival overgrowth patients, and the intensity of inhibition is related to the degree of gingival hyperplasia and drug concentration, while the intervention may relate to the decrease of TGF- β1.
