Cytocompatibility of injectable bioactive glass/calcium sulfate composite bone implants

摘要 目的 探讨生物活性玻璃/硫酸钙(PSC/CS)骨水泥的细胞相容性.方法 通过复合PSC[10.8%五氧化二磷(P2O5)与54.2%二氧化硅(SiO2)、35%氧化钙(CaO)]与CS得到复合材料(PSC/CS),用噻唑蓝(MTT)法测定PSC/CS与硫酸钙骨水泥(CSC)浸提液对骨髓间充质干细胞(BMSCs)增殖的影响,并进行细胞毒性分级,同时检测PSC/CS和CSC BMSCs黏附的影响,比色法检测各组碱性磷酸酶(ALP)活性.结果 增殖结果显示,在第3天(PSC/CS组:0.328±0.002、CSC组:0.312±0.013)、第6天(PSC/CS组:0.708±0.017、CSC组:0.658±0.021)、第9天(PSC/CS组:1.123±0.124、CSC组:0.933±0.028),差异有统计学意义(P =0.103、0.075、0.061);细胞相对增殖率在第3天分别为98.34%和97.65%,第6天分别为98.27%和95.36%,第9天分别为97.24%和94.34%,细胞毒性分级均为Ⅰ级,细胞黏附率(P =0.047、0.034、0.032)及ALP活性测定结果(P=0.042、0.043、0.026)实验组明显高于对照组,差异有统计学意义.结论 含PSC的PSC/CS具有良好的细胞相容性. Objective To investigate the cytocompatibility of bioactive glass/calcium sulfate cement.Methods The composites (PSC/CS) were obtained by complex bioactive glass (10.8% P2O5 and 54.2% SiO2,35% CaO,PSC) and calcium sulfate (CS).The effects of PSC/CS and calcium sulfate cement (CSC) extracts on the proliferation of bone marrow mesenchymal stem cells (BMSCs) were measured by methyl thiazol tetrazolium (MTT) method,and the cytotoxicity was graded.Meanwhile,the adhesion of PSC/CS and CSCs was detected,and the alkaline phosphatase activity of each group was detected by colorimetry.Results The proliferation results (Tbe P value is:0.103,0.075,0.061,respectively) showed that on the 3rd day (PSC/CS group:0.328 ± 0.002,CSC group:0.312 ± 0.013),on the 6th day (PSC/CS group 0.708 ±0.017,CSC group 0.658 ±0.021),on the 9th day (PSC/CS group:1.123 ±0.124,and CSC group:0.933 ±0.028).Tbe relative cell proliferation rates were 98.34% and 97.65% on day 3,98.27% and 95.36% on day 6,and 97.24% and 94.34% on day 9,respectively,Cytotoxieity grade were grade Ⅰ,cell adhesion rate (The P value is:0.047,0.034,0.032,respectively) and alkaline phosphatase activity of the experimental group (The P value is:0.042,0.043,0.026,respectively) was significantly higher than the control group,the difference was statistically significant.Conclusion PSC/CS with bioactive glass has a good cytocompatibility.
