摘要
目的初探百合的甲醇提取物、百合总生物碱对人胃癌SGC-7901细胞增殖和凋亡的影响。方法不同浓度的百合甲醇提取物、百合总生物碱作用SGC-7901细胞,MTT法检测SGC-7901细胞的生长抑制率;荧光显微镜观察SGC-7901细胞的形态变化;流式细胞仪检测SGC-7901细胞周期改变及其凋亡率。结果随着百合甲醇提取物、百合总生物碱作用SGC-7901细胞的时间、浓度递增,抑制SGC-7901细胞增殖的作用越明显,百合甲醇提取物最大浓度的抑制率可达到98.32%;百合总生物碱最大浓度的抑制率可达到91.93%,细胞逐渐失去正常的细胞形态,细胞边缘不整齐,细胞发生皱缩;阻滞细胞周期于G2/M期;可以诱导细胞的凋亡。结论百合甲醇提取物、百合总生物碱阻滞SGC-7901细胞于G2/M期,诱导SGC-7901细胞凋亡是抑制SGC-7901细胞增殖的主要机制之一。
Objective The present study is to investigate the effect of carbinol extracts and alkaloids from Lilium brownii var. viridulum on the proliferation and apoptosis of SGC-7901 cells. Methods SGC-7901 cells were incubated with different concentrations of carbinol extract or alkaloid. MTT assay was used to measure the inhibition rate of SGC-7901 cell proliferation. Fluorescence microscopes were used to observe morphological changes of SGC-7901 cells. Flow cytometer was employed to detect cell cycle and apoptosis of SGC-7901 cells. Results Inhibition of SGC-7901 cell proliferation was enhanced as the increase of drug concentrations and the elapse of time. Treatment with alkaloid or carbinol extract deteriorated the morphology of SGC-7901 cells in a dose-dependent manner. Alkaloid or carbinol extract arrested SGC-7901 cells in G2/M phase. Treatment with alkaloid or carbinol extract induced apoptosis of SGC-7901 cells in a dose-dependent manner. Conclusion The present study demonstrates that alkaloids and carbinol extracts from Lilium brownii var. viridulum inhibit the proliferation of gastric carcinoma SGC-7901 cells by arresting the cells in G2/M phase. The induction of SGC-7901 cell apoptosis may be an important mechanism.
