树突状细胞基因修饰疗法在1型糖尿病治疗中的应用研究

摘要 树突状细胞(dendritic cells,DC)是已知功能最强的专职抗原提呈细胞(antigen presenting cell ,APC),其最大特点是刺激初始T细胞(naive T cells)的活化与增殖。DCs不仅在机体抗感染、肿瘤等免疫应答过程发挥作用,同时维持中枢耐受和外周耐受,防止自身免疫病的发生发展。Ⅰ型糖尿病(type 1 diabetes ,T1D)是一种常见的自身免疫病,由免疫耐受异常所致,其治疗存在巨大挑战。DCs中多种基因表达和功能异常与T1D密切相关,因此人们尝试修饰DCs相关基因治疗T1D。 Dendritic cells (DCs) are known as the most potent professional antigen presenting cell (APC). The most important feature of DCs is to stimulate the activation and proliferation of the na?ve T cells.DCs play a unique role in the immune system, which participate not only in anti-infection, anti-tumor and other immune response process of the organism, but also in the maintenance of central tolerance and peripheral tolerance to prevent the development of autoimmune diseases.Type 1 diabetes (T1D) is a common autoimmune disease, triggered by dysfunction of immune tolerance.While the of T1D treatment is still a huge challenge.Since the abnormality of various gene expression and function in DCs are closely related with T1D, scientists try to treat T1D by modifying the DCs-related gene.
