The role of autophagy in cisplatin resistance of tumor

摘要 肿瘤是现社会致死率最高的疾病之一,化疗仍是其最广泛且有效的治疗手段,顺铂作为一种细胞毒药物被应用于临床恶性肿瘤的化疗,但是几乎所有的化疗最终会因耐药而导致治疗失败。自噬是一种精细调节的通路,主要是指胞浆中细胞器或其他组分经自噬溶酶体降解的过程。自噬与多种疾病有着紧密的联系。众多的研究证明,癌细胞接受化疗意味着自噬的激活,而自噬对肿瘤细胞的保护机制介导了肿瘤耐药的发生,但自噬在肿瘤的顺铂耐药中的机制仍待阐明。因此进一步研究自噬在肿瘤顺铂耐药中的机制以及寻找新的逆转耐药靶点是科研界面临的主要问题。 Malignant tumor is one of the most fatal diseases in modern society, which chemotherapy is still the most extensive and effective clinical treatment.Cisplatin, a cytotoxic drug, has been used for the clinical chemotherapy of malignant tumors, while its therapeutic effects always end of failure due to drug resistance.Autophagy is a fine control pathway, closely related to many diseases, mainly characterized by the degradation of organelles and other components in cytoplasm by autolysosome.Many studies proved that autophagy is activated while chemotherapy, indicating that autophagy protects tumor cells that maybe inducing tumor resistance, while the mechanism of cisplatin resistance by autophagy is remaining unknown.Thus, further researches need to be done for the mechanisms of cisplatin resistance mediated by autophagy and we should find new targets to reverse drug resistance.
