摘要
目的探讨结肠癌细胞株HCT116皮下种植瘤的组织细胞再行尾静脉注射成瘤的可行性。方法将30只裸鼠随机分为A、B、C组,每组10只。A组尾静脉注射生理盐水,B组尾静脉注射HCT116细胞悬液0.1 ml,C组尾静脉注射HCT116皮下种植瘤组织悬液0.1 ml。将HCT116细胞皮下种植瘤组织悬液离心分为细胞与组织悬液。另取20只裸鼠随机分为细胞组和悬液组(n=10),尾静脉注射细胞或悬液。观察各组裸鼠成瘤及转移情况。结果 A组无肿瘤形成。B组裸鼠尾静脉注射HCT116细胞成功9只,8只形成肺部转移瘤,其中2只同时形成肝部转移瘤。C组裸鼠尾静脉注射HCT116皮下种植瘤组织悬液成功9只,初期有4只尾根部出现肿块,30天后5只裸鼠的骶部、颈部出现肿瘤(严重者有1只同时胸窝皮下也形成肿瘤),50天后形成肺部转移瘤,死亡;未形成皮下种植瘤的4只裸鼠中有3只形成肺部转移瘤后死亡。细胞组裸鼠注射离心后的细胞成功8只,仅1只形成肺部转移瘤。悬液组裸鼠注射离心后组织悬液无肿瘤形成。结论 HCT116皮下种植瘤组织悬液尾静脉注射后,能定向地从血管迁移至裸鼠皮下组织成瘤。
Objective To investigate the feasibility of forming metastatic tumor by tail intravenous injection of colon cancer HCT116 cells from subcutaneous implants. Methods Thirty nude mice were randomly divided into A,B and C groups,with 10 mice in each group. In group A,mice were given normal saline by tail vein injection; in group B,mice were given HCT116 cell suspension0. 1 ml by tail vein injection; mice in group C were injected with subcutaneous implants suspension 0. 1 ml via tail vein. HCT116 cells subcutaneous implants suspension was centrifuged into cells and tissue fluid. Another 20 mice were randomly divided into cell group( n= 10) and fluid group( n = 10) receiving injection of cells and fluid by tail rein. The tumor formation and metastasis were observed in each group. Results Mice in group A were without tumor formation. Injection of HCT116 cells in group B was successful in 9 nude mice. Only 8 formed lung metastases and 2 of them formed hepatic metastatic tumor spontaneously. Injection of HCT116 tissue suspension in group C via tail vein was successful in 9 nude mice. Initially,masses were found at caudal root in 4 mice,and masses were observed at sacral and neck in 5 mice after 30 days,among which subcutaneous mass of chest was found in 1 mouse. Lung metastases formed after 50 days and then the 5 mice died. Three of the 4 nude mice without subcutaneous tumors died after the formation of lung metastases. In cell group,8 nude mice were successfully injected with centrifuged cells,and lung metastases were formed in 1 mouse.Mice in fluid group injected with tissue fluid were without tumor formation. Conclusion HCT116 can migrate from the blood vessel to the subcutaneous tissue of nude mice after injection of subcutaneous implants suspension.
