摘要
通过酶标比浊法、试剂盒法和邻硝基苯B—D半乳吡喃糖苷(0NPG)法测定姜黄素对致病性金黄色葡萄球菌(Staphylococcus aureus)生长曲线、细胞壁及细胞膜渗透性的影响。同时,构建大鼠子宫内膜炎模型,用ELISA试剂盒检测各试验组中白介素6(IL-6)、白介素8(IL-8)和肿瘤坏死因子-α(TNF-α)的变化情况来监测治疗效果。结果显示:(1)姜黄素对致病性S.aureus的生长曲线、细胞壁和细胞膜渗透性均有影响,因而起到抑菌的效果。当姜黄素作用于菌体细胞后,使细菌细胞壁被损伤,从而使细胞膜的通透性增加,细胞结构被破坏,从而起到抑菌的效果。(2)姜黄素中、高剂量组和环丙沙星组能显著降低患子宫内膜炎大鼠血清中的IL-6、IL-8和TNF-α的浓度,而模型组这些炎性介质的浓度不断的升高。结果表明:姜黄素的抑菌效果最佳,优于环丙沙星;姜黄素能显著抑制子宫内膜炎大鼠血清中IL-6、IL-8和TNF—α的表达来发挥调控子宫内环境动态平衡。
The study was conducted to investigate the antibacterial mechanism of curcumin on the pathogenic Staphylococcus aureus and its on anti-inflammatory activity in serum of rats with endometritis. The effects of curcumin on the growth curve, the hydrophobicity, the cytoderm and cell membrane of S. aureus were determined by turbidimetry,the microbial adhesion to solvents method,reagent kit and ONPG as a substrate,respectively. Moreover,an animal model was established by S. aureus, and the levels of IL-6, IL-8 and TNF-α were detected by ELISA kit. The results showed that the curcumin on the growth curve,bacterial cell surface hydrophobicity, cytoderm and cell membrane permeability of pathogenic S. aureus had an effect, so that the inhibitory effect. After curcumin effects on bacteria cell,its surface hydrophobicity was changed and the integrity of the cell wall was destroyed gradually, and then the permeability of cell membrane increased, lead to destruction of the cell structure that was antimicrobial mechanism of curcumin. The levels of IL-6, IL-8 and TNF-α in ciprofloxacin group and high dose group of curcumin reduced,but the concentration of these inflammatory mediators increased in the model group. Curcumin has the most obvious effect,and the effects is better than that of the antibiotic ciprofloxacin,and inhibited the expression of IL-6,IL-8 and TNF-α.
