蒲公英提取物对佐剂性关节炎大鼠的保护作用及机制 被引量:16

Protective effect and mechanism of dandelion extract on adjuvant arthritis in rats

摘要 采用弗氏完全佐剂(FCA)建立大鼠佐剂性关节炎模型,设空白组、模型组、阳性组和蒲公英提取物高、中、低剂量组(10.0,5.0,2.5g/kg),分别对大鼠足趾肿胀、体质量、脾脏指数、胸腺指数和血清中TNF-α、IL-1β、PGE2、OPG、RANKL的含量进行测定并计算RANKL/OPG,HE染色观察大鼠踝关节软骨组织及滑膜组织的病理变化。结果显示:蒲公英提取物高、中、低剂量组均不同程度降低佐剂性关节炎大鼠原发性和继发性足趾肿胀,增加体质量,下调脾脏指数和胸腺指数,抑制血清中TNF—α、IL-1β、PGE2、RANKL的分泌,促进OPG的分泌,抑制大鼠踝关节炎性细胞浸润和滑膜增生。结果表明:蒲公英提取物对大鼠佐剂性关节炎具有一定的保护作用,为临床应用提供有效的依据。 In order to study the protective effect and mechanism of dandelion extract on adjuvant arthritis in rats,the model of adjuvant arthritis was established by Freund's complete adjuvant. The rats were divided into blank group,model group, positive group and dandelion extract high, medium and low dose groups(10.0,5.0,2.5 g/kg), ankle swelling, body weight, spleen index and thymus index were measured,the contents of TNF-α, IL-113,PGE2, OPG, RANKL in serum were determined and the value of RANKL/OPG was calculated. The results showed that dandelion extract high,medium and low dose groups inhibited the degree of the primary swelling and second swelling,increased the body weight,decreased the spleen index and thymus index, inhibited the secretion of TNF-α, IL-113, PGE2, RANKL and promoted the secretion of OPG in serum, reduced the in- filtration of inflammation cell and synovial hyperplasia. It suggests that dandelion extract has a certain protective effect on adjuvant arthritis in rats,which provides an effective basis for its clinical application.
