芪玉三龙汤对肺癌p70S6激酶和真核翻译起始因子4E的影响 被引量:4

摘要 目的:研究芪玉三龙汤对肺癌生长的关键合成蛋白p70S6激酶(p70S6K)和真核翻译起始因子4E(eIF4E)表达的影响。方法:基于前期研究基础,采用LLC细胞培养移植法构建移植肺癌模型,荷瘤成功后随机分为模型组(M),化疗组(C),中药组(低、中、高剂量组,分别命名为QL、QM、QH),联合组(CQH),低、中、高剂量组小鼠分别按(20.12、40.24、80.48)g/(kg·d)灌胃给药,化疗组以0.4 mL顺铂腹腔注射给药,联合组以中药高剂量灌胃和顺铂腹腔注射联合给药,模型组以等量生理盐水给药,1次/d,连续给药10 d;称取瘤质量,计算抑瘤率;透射电镜观察肿瘤细胞超微结构的变化;RT-qPCR法检测Ras mRNA表达水平;Western Blot法检测肿瘤组织eIF4E、p-eIF4E、p70S6K、p-p70S6K蛋白表达。结果:芪玉三龙汤可抑制肺癌移植瘤生长,电镜下可见凋亡小体,用药各组均可下调Ras转录水平,但中药各组之间无明显差异(P〉0.05),联合组与化疗组之间相比无统计学意义(P〉0.05)。同时,该复方均可降低肿瘤组织eIF4E、p-eIF4E、p70S6K、p-p70S6K蛋白表达。结论:芪玉三龙汤可抑制肺癌生长,诱导其镜下凋亡,与下调肺癌生长的关键合成蛋白p70S6K和eIF4E表达相关。 Objective:To observe the effects of Qiyu Sanlong Decoction(QYSL) on expressions of p70S6 K and eIF4 E in lung cancer. Methods:The model of lung cancer subcutaneous allografts was established in C57 mice using Lewis lung carcinoma cells line(LLC). The mice were randomly divided into six groups:model group,chemotherapy group,QYSL groups(low dosage,middle dosage,high dosage) and combination group. QL,QM and QH were fed with QYSL according to 20. 12 g/(kg·d),40. 24 g/(kg·d),80. 48 g/(kg·d). C were given 0. 4 mL cisplatin by intraperitoneal injecting.CQH were administered with high dosage of QYSL and cisplatin. M were administered with the same amount of saline,once a day,lasting for 10 d. The tumor was weighed and tumor inhibition rates were calculated. Transmission electron microscope observed the ultrastructural changes of tumor cells to determine whether QYSL can induce apoptosis. RT-qPCR measured the expression of Ras mRNA. Western Blot measured the expression levels of eIF4 E and p70 S6 K in tumor. Results:QYSL could inhibit the growth of tumor. Apoptotic bodies were seen under electron microscope. The treatment group could down-regulate the transcription level of ras,but there is no significant difference between the Chinese medicine group(P〉0. 05). Compared with chemotherapy group,combination group had no statistical significance(P〉0. 05). At the same time,QYSL could reduce the eIF4 E,p-eIF4 E,p70 S6 K,p-p70S6 K expressions of tumor tissue. Conclusion:QYSL could inhibit tumor growth and induce apoptosis,which was associated with down-regulation of synthetic protein p70 S6 K and eIF4 E expressions in tumor tissue.
